Forskolin is a naturally occurring bioactive compound extracted from the root of Coleus forskohlii, a plant belonging to the mint family (Lamiaceae). Native to South and Southeast Asia and parts of Africa, this plant has been used in traditional Ayurvedic medicine for centuries and is now the subject of extensive scientific research.
The primary pharmacological action of forskolin is the activation of the enzyme adenylyl cyclase. This enzyme converts ATP (adenosine triphosphate) into cAMP (cyclic adenosine monophosphate), an important intracellular second messenger. Elevated cAMP levels influence a wide range of cellular processes, including:
Forskolin is used in some countries as eye drops to reduce intraocular pressure in patients with glaucoma. Clinical studies have demonstrated its efficacy in lowering eye pressure.
Due to its vasodilatory and cardiotonic properties, forskolin has been investigated in the treatment of heart failure. Intravenous administration has shown positive effects on cardiac function.
By relaxing bronchial smooth muscle, forskolin can act as a bronchodilator and has been studied in the context of bronchial asthma.
Forskolin is widely marketed as a dietary supplement to support weight management and promote lean body mass. However, the scientific evidence for these uses remains limited. Some smaller studies suggest a potential promotion of lipolysis and a modest increase in testosterone levels, but large-scale, well-controlled human trials are still lacking.
In clinical studies, dosages of 25 to 250 mg of forskolin per day (usually as a standardized extract containing 10–20% forskolin) have been commonly used. The appropriate dosage depends on the indication and should always be determined in consultation with a qualified healthcare professional.
Forskolin is generally considered safe when taken orally in moderate amounts. Possible side effects include:
Caution is advised for individuals taking antihypertensive medications, blood thinners, or other cardiac drugs, as interactions are possible. Pregnant and breastfeeding women should not take forskolin without medical advice.
Forskolin may interact with several medications, including:
