Oleoylethanolamide (abbreviated OEA) is a naturally occurring bioactive lipid belonging to the family of fatty acid ethanolamides. It is produced primarily in the cells lining the small intestine in response to dietary fat intake, especially oleic acid – the main fatty acid found in olive oil. OEA acts as an endogenous signaling molecule that helps regulate appetite, energy metabolism, and lipid balance.
OEA exerts its effects mainly through the peroxisome proliferator-activated receptor alpha (PPAR-α), a nuclear receptor expressed in the intestine, liver, and other metabolically active tissues. Activation of PPAR-α by OEA triggers several important responses:
OEA also activates the GPR119 receptor (G protein-coupled receptor 119), which stimulates the release of incretin hormones such as GLP-1, thereby influencing insulin secretion and glucose metabolism.
Within the human body, OEA fulfills several important physiological roles:
Because of its role in appetite regulation and fat metabolism, OEA is used as an active ingredient in various dietary supplements marketed for weight management. Typical supplement dosages range from 100 to 300 mg per day, although the optimal dose has not yet been conclusively determined by clinical trials.
Supplements are usually taken before meals to complement the natural satiety signal that occurs after food consumption.
The current body of evidence on OEA is promising but largely based on animal and in vitro studies. Clinical human trials demonstrating clear efficacy in obesity or metabolic disorders remain limited. OEA is generally considered well tolerated; serious adverse effects have not been reported, though mild gastrointestinal complaints such as nausea have occasionally been noted.
OEA is rapidly broken down in the body by the enzyme fatty acid amide hydrolase (FAAH). Since OEA acts via PPAR-α, theoretical interactions with other PPAR-α agonists (e.g., fibrates or certain lipid-lowering drugs) cannot be ruled out. Individuals taking medications or with existing health conditions should consult a physician before using OEA-containing supplements. Insufficient safety data are available for pregnant or breastfeeding women, and use during these periods is therefore not recommended.
