Formononetin is a naturally occurring isoflavone – a subclass of phytoestrogens – found primarily in plants such as red clover (Trifolium pratense), soy, licorice root, and other legumes. It belongs to the broader class of polyphenols and is synthesized as a secondary metabolite in these plants. After ingestion, formononetin is metabolically converted in the human body to equol, a biologically active compound with estrogenic activity.
Formononetin exerts its effects primarily through binding to estrogen receptors, with a particular affinity for the beta subtype (β). This binding occurs with considerably lower potency than that of endogenous estrogen (17β-estradiol), which is why formononetin is classified as a weak phytoestrogen.
