What Is a Soft Gelatin Capsule?
A soft gelatin capsule – also known as a softgel or soft capsule – is a pharmaceutical dosage form consisting of a flexible, hermetically sealed outer shell and a liquid or semi-solid inner filling. The shell is traditionally made from animal-derived gelatin, although plant-based alternatives such as hydroxypropyl methylcellulose (HPMC) are increasingly used to meet vegetarian and vegan requirements. Soft gelatin capsules are widely used for both prescription medications and over-the-counter dietary supplements.
Structure and Manufacturing
Soft gelatin capsules consist of two main components:
- Capsule shell: A flexible, airtight outer layer made from gelatin, starch, or HPMC that protects the contents from the environment.
- Fill material: A liquid, oil-based solution, emulsion, suspension, or semi-solid mass in which the active ingredient is dissolved or dispersed.
They are typically produced using a rotary die encapsulation process, in which two ribbons of gelatin are simultaneously shaped, filled, and sealed, forming a seamless, uniform capsule.

Soft gelatin capsules offer several advantages over other dosage forms:
- Enhanced bioavailability: Fat-soluble active ingredients dissolved in an oily carrier are more readily absorbed in the gastrointestinal tract.

Soft gelatin capsules are used for a broad range of products, including:
- Fish oil and omega-3 fatty acids (e.g., for cardiovascular health support)
- Fat-soluble vitamins such as Vitamin D, Vitamin E, and Vitamin A
- Plant-based oils such as flaxseed oil, evening primrose oil, or garlic oil
- Pharmaceutical drugs such as ibuprofen soft capsules, immunosuppressants (e.g., ciclosporin), and antiretrovirals
- Probiotics and enzyme preparations
Soft Capsules vs. Hard Capsules
Unlike hard capsules (also called two-piece capsules), which consist of two separate rigid halves filled with powder or granules, soft gelatin capsules are manufactured as a single sealed unit and are specifically designed for liquid or semi-solid fills. This makes them particularly suitable for oil-based or poorly water-soluble active ingredients that benefit from being dissolved in a lipid vehicle.
Instructions for Use
Soft gelatin capsules should always be swallowed whole and unchewed with a sufficient amount of water. Breaking or chewing the capsule can damage the shell and release the contents prematurely, potentially reducing effectiveness or causing irritation. Some preparations are better tolerated when taken with food, which may also enhance absorption. Always refer to the package insert or consult a healthcare professional or pharmacist if in doubt.
