What is Allergen Characterization?

Allergen characterization is a scientific process in which substances capable of triggering allergic reactions are systematically analyzed and described. The aim is to precisely understand the molecular structure, biochemical properties, and immunological potential of an allergen. This forms the foundation for the diagnosis, treatment, and prevention of allergic diseases.

What Are Allergens?

Allergens are substances – usually proteins or glycoproteins – that can trigger an exaggerated immune response (allergy) in genetically predisposed individuals. They are found in a wide variety of sources, including:

  • Pollen (e.g., birch, grasses, mugwort)
  • Animal-derived products (e.g., house dust mites, animal dander, insect venom)
  • Foods (e.g., peanuts, milk, wheat flour)
  • Molds and yeasts
  • Medications and chemical compounds

Methods of Allergen Characterization

Various scientific methods are used to characterize allergens:

Biochemical Analysis

Techniques such as SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis) and immunoblotting are used to separate and identify protein components within an allergen mixture. These methods reveal which proteins are recognized by the IgE antibodies of sensitized patients.

Molecular Biology Methods

Using recombinant DNA technology, individual allergen molecules can be produced in large quantities, isolated, and precisely analyzed. Recombinant allergens enable standardized and reproducible diagnostics and support the development of targeted immunotherapies.

Mass Spectrometry

Mass spectrometry allows the precise determination of the molecular mass and structure of an allergen and helps identify protein families and cross-reactivities between different allergens.

Crystallographic and Structural Analyses

Techniques such as X-ray crystallography or nuclear magnetic resonance (NMR) spectroscopy can reveal the three-dimensional structure of an allergen. This is essential for understanding which regions of the protein – known as epitopes – are recognized by the immune system.

Classification of Allergens

Allergens are named internationally according to a standardized nomenclature system established by the WHO/IUIS Allergen Nomenclature Sub-Committee. The name consists of an abbreviation of the genus and species of the source organism followed by a sequential number (e.g., Bet v 1 for the major allergen of birch, Betula verrucosa).

Allergens are also classified by their clinical significance:

  • Major allergens: Recognized by more than 50% of sensitized patients.
  • Minor allergens: Recognized by fewer than 50% of sensitized patients.

Cross-Reactivity

A key concept in allergen characterization is cross-reactivity. This occurs when the immune system reacts to structurally similar proteins from different sources. A well-known example is the birch-mugwort-celery syndrome, in which patients with a birch pollen allergy may also react to certain vegetables and fruits because their proteins share structural similarities with Bet v 1.

Clinical Significance

The precise characterization of allergens has far-reaching implications for clinical practice:

  • Improved diagnostics: Component-resolved allergy diagnostics (also called molecular allergy diagnostics) allow for more precise identification of the causative allergens.
  • Allergen-specific immunotherapy (AIT): Standardized allergen extracts and recombinant allergens enable more targeted and safer desensitization treatments.
  • Risk assessment: Certain allergen components, such as Ara h 2 in peanut allergy, are associated with an increased risk of severe anaphylactic reactions.
  • Labeling requirements: In the food industry, allergen characterization forms the basis for legally required allergen labeling.

Allergen Characterization in Research

Current research increasingly uses bioinformatics tools and artificial intelligence to predict allergenic proteins in newly developed foods, genetically modified organisms, or novel substances. International databases such as AllerHunter, SDAP (Structural Database of Allergenic Proteins), and the WHO/IUIS Allergen Database provide comprehensive information on characterized allergens.
