Amelogenin is an extracellular matrix protein produced by ameloblasts – the enamel-forming cells – during tooth development. It belongs to the group of enamel matrix proteins and is essential for the mineralisation and structural organisation of tooth enamel, the hardest biological tissue in the human body.
Amelogenin accounts for approximately 90% of all proteins in the developing enamel matrix. It regulates the growth and orientation of hydroxyapatite crystals, which give tooth enamel its characteristic hardness and resilience.
Mutations in the AMELX gene can cause amelogenesis imperfecta, a hereditary condition in which the tooth enamel is incomplete, thin, or poorly mineralised. Affected individuals experience increased sensitivity, discolouration, and a higher susceptibility to dental caries.
Enamel matrix derivatives (EMD), composed mainly of amelogenin and derived from the enamel organ of young pigs, are used in regenerative periodontal therapy. Products such as Emdogain® promote the regeneration of gingival tissue, cementum, and alveolar bone in patients with periodontitis.
Because the AMELX and AMELY genes are located on different sex chromosomes, amelogenin is widely used in forensic genetics for sex determination from biological trace evidence such as teeth and blood. The difference in PCR amplicon length allows reliable distinction between male (XY) and female (XX) genetic material.
Analysis of the amelogenin locus is a standard component of DNA profiling (e.g. STR analysis) and is used in both forensic medicine and prenatal diagnostics. Rare deletions in the AMELX gene region may lead to misidentification of genetic sex.
