Amitriptyline is a medication belonging to the class of tricyclic antidepressants (TCAs). It has been used in clinical medicine since the 1960s and remains one of the most widely prescribed antidepressants worldwide. In addition to treating depression, amitriptyline is commonly used today for chronic pain management and certain sleep disorders.
Amitriptyline works by inhibiting the reuptake of the neurotransmitters serotonin and noradrenaline (norepinephrine) in the nerve cells of the brain. This means these chemical messengers remain available in the synaptic cleft for longer, improving communication between nerve cells. This mechanism underlies both its mood-lifting and pain-relieving effects.
Amitriptyline also has antihistaminergic (sedating) and anticholinergic properties, which can be therapeutically beneficial but also contribute to certain side effects.
