AOD-9604 (Anti-Obesity Drug 9604) is a synthetic peptide derived from the C-terminal fragment of human growth hormone (hGH), specifically encompassing amino acids 177 to 191. It was originally developed as a potential treatment for obesity. Unlike the full-length growth hormone, AOD-9604 does not affect insulin levels or stimulate cell growth; instead, it specifically targets fat metabolism.

Mechanism of Action

AOD-9604 mimics the lipolytic (fat-breaking) properties of natural growth hormone without triggering its anabolic (tissue-building) effects. The key mechanisms include:

  • Lipolysis: AOD-9604 stimulates the breakdown of stored fat cells (triglycerides) into free fatty acids, which are then used as an energy source.
  • Inhibition of Lipogenesis: The peptide inhibits the formation of new fat tissue from carbohydrates and other nutrients.
  • Beta-3 Adrenergic Receptors: AOD-9604 interacts with beta-3 adrenergic receptors in adipose tissue, further promoting fat breakdown.
  • No IGF-1 Stimulation: Unlike full-length growth hormone, AOD-9604 does not stimulate the production of Insulin-like Growth Factor 1 (IGF-1), thereby avoiding unwanted growth-related side effects.

Medical Research and Applications

AOD-9604 was originally developed by the Australian pharmaceutical company Metabolic Pharmaceuticals and investigated in clinical trials for its potential to treat obesity. Preclinical studies demonstrated promising results in reducing body fat. However, Phase II and Phase III clinical trials in humans did not provide sufficient evidence of significant weight reduction, and the peptide has not received regulatory approval as an anti-obesity drug to date.

Beyond weight management, AOD-9604 is being explored for additional potential therapeutic applications, including:

  • Cartilage Regeneration: Some preclinical studies suggest a possible role in repairing cartilage tissue, making it of interest for osteoarthritis treatment.

Usage and Dosage

AOD-9604 is not available as an approved pharmaceutical drug and is therefore not officially recommended for medical treatment.
