What is CJC-1295?

CJC-1295 is a synthetically manufactured peptide belonging to the class of Growth Hormone Releasing Hormone (GHRH) analogues. It was originally developed to increase the secretion of growth hormone (GH) from the pituitary gland. CJC-1295 is a modified version of the naturally occurring Growth Hormone Releasing Hormone (GHRH), which is produced in the hypothalamus.

The peptide was developed by the Canadian biotechnology company ConjuChem and is also known as DAC:GRF (Drug Affinity Complex: Growth Hormone Releasing Factor). It is not approved as a pharmaceutical drug and is currently not recommended for clinical use in humans. Nevertheless, it is widely -- though illegally -- used in competitive sports and anti-aging communities.

Mechanism of Action

CJC-1295 binds to the GHRH receptor in the pituitary gland, mimicking the action of natural GHRH. This leads to increased secretion of growth hormone into the bloodstream. Compared to native GHRH, which has a very short half-life of only a few minutes, CJC-1295 has a significantly extended duration of action.

This is achieved through two mechanisms:

  • Albumin binding: Through a special chemical modification (DAC technology), CJC-1295 binds to the plasma protein albumin in the blood. This protects the peptide from enzymatic degradation and extends its half-life to several days.
  • Peptide structure stabilisation: Specific amino acid substitutions protect the molecule from degradation by Dipeptidyl Peptidase IV (DPP-IV).

The result is a sustained, pulsatile release of growth hormone, leading to an increase in Insulin-like Growth Factor 1 (IGF-1) in the liver and other tissues. IGF-1 mediates many of the anabolic and growth-promoting effects of growth hormone.

Areas of Application (Research Context)

In scientific research, CJC-1295 has primarily been investigated as a potential therapy for the following conditions:

  • Growth hormone deficiency: In adults and children with clinically relevant GH deficiency.
  • Muscle wasting (sarcopenia): Related to the natural age-related decline in muscle mass.
  • Lipid metabolism disorders: As growth hormone promotes fat breakdown (lipolysis).
  • Anti-aging research: To investigate the effects of increased GH secretion on the aging process.

It is important to emphasise that CJC-1295 has not yet been sufficiently investigated in any phase of clinical trials to obtain market approval. Most studies involved small patient groups and short observation periods.

Illegal Use in Sports and Anti-Aging

Despite lacking regulatory approval, CJC-1295 is widely used by bodybuilders, competitive athletes, and within the anti-aging community. The hoped-for effects include:

  • Increased muscle mass and strength
  • Reduction of body fat
  • Improved recovery after training
  • Improvement in skin quality and sleep quality

CJC-1295 is classified as a prohibited substance by the World Anti-Doping Agency (WADA) and is listed on the Prohibited List. Its use constitutes a doping violation.

Side Effects and Risks

Since CJC-1295 is not officially approved, long-term safety data are limited. The following side effects are known from clinical studies and reports:

  • Local reactions: Redness, swelling, or pain at the injection site
  • Water retention (oedema): Due to the anabolic effects of growth hormone
  • Carpal tunnel syndrome: Caused by fluid retention in the wrist area
  • Blood pressure changes
  • Headaches and dizziness
  • Hypoglycaemia: Possible interaction with insulin metabolism
  • Increased cancer risk: Chronically elevated IGF-1 levels are associated with an increased risk of certain cancers (e.g., colorectal, breast, and prostate cancer).
  • Acromegaly-like symptoms: With excessive, sustained growth hormone levels (coarsening of facial features, joint pain)

Legal Status

CJC-1295 is not approved as a pharmaceutical drug in most countries, including Germany, Austria, and Switzerland. It may neither be sold nor marketed as a medicinal product. Possession and acquisition may fall under national pharmaceutical law or anti-doping legislation, depending on the country. In competitive sport, it is explicitly prohibited by WADA.
