What Is a Defense Protein?

A defense protein is a protein produced by the immune system to protect the body against infections, foreign substances, and disease-causing microorganisms. Defense proteins play a central role in both the innate and adaptive immune response and are essential for human health.

Types of Defense Proteins

Antibodies (Immunoglobulins)

Antibodies are Y-shaped proteins produced by B lymphocytes (a type of white blood cell). They recognize specific structures on the surface of pathogens, known as antigens, and bind to them. This marks the pathogens for destruction by other immune cells or neutralizes them directly. There are five classes of antibodies: IgA, IgD, IgE, IgG, and IgM.

Complement Proteins

The complement system consists of more than 30 proteins found in blood plasma that are activated in a cascade-like manner. They can directly destroy pathogens, trigger inflammatory responses, and recruit other immune cells. The complement system is an important part of innate immunity.

Acute-Phase Proteins

Acute-phase proteins are increasingly produced by the liver during inflammation and infection. Well-known examples include C-reactive protein (CRP) and serum amyloid A. They support the immune response by regulating inflammatory processes and binding pathogens.

Interferons

Interferons are signaling proteins released by infected cells to warn neighboring cells of a viral infection. They inhibit the replication of viruses and activate immune cells such as natural killer cells.

Defensins

Defensins are small antimicrobial peptides found in many body tissues, including the skin, mucous membranes, and white blood cells. They act directly against bacteria, fungi, and viruses by disrupting their cell membranes.

Function and Importance

Defense proteins fulfill numerous roles within the immune system:

  • Recognition and neutralization of pathogens
  • Marking of pathogens for destruction by immune cells
  • Regulation of inflammatory responses
  • Protection of body cells from viral infection
  • Direct antimicrobial activity

Clinical Significance

A deficiency in certain defense proteins can lead to increased susceptibility to infections. For example, an antibody deficiency (agammaglobulinemia or hypogammaglobulinemia) causes recurrent bacterial infections. Disorders of the complement system are also associated with conditions such as systemic lupus erythematosus and recurrent meningitis infections.

Conversely, excessive activity of defense proteins can contribute to autoimmune diseases or allergies, when the immune system mistakenly targets the body own structures or harmless substances.

Diagnostics

In clinical practice, defense proteins are frequently measured in laboratory tests to detect diseases or monitor the course of immune reactions. Common laboratory values include:

  • Immunoglobulins (IgA, IgG, IgM, IgE) in blood serum
  • C-reactive protein (CRP) as an inflammatory marker
  • Complement factors (e.g., C3, C4)
