What is Dihexa?
Dihexa is a synthetic, low-molecular-weight peptide derivative originally developed at Washington State University. It is derived from Angiotensin IV (Ang IV), a peptide fragment of the renin-angiotensin system. Dihexa was specifically engineered to readily cross the blood-brain barrier and exert its effects within the central nervous system. It belongs to the class of nootropic compounds — substances intended to support cognitive functions such as memory, learning, and mental performance.
Mechanism of Action
The primary mechanism of action of Dihexa involves activation of the Hepatocyte Growth Factor receptor (HGF/c-Met) signaling pathway. Dihexa acts as a potentiating ligand that enhances HGF signaling, thereby stimulating synaptogenesis — the formation of new neuronal connections (synapses). This activity is believed to promote neural plasticity and thereby support learning and memory processes.
- Binding to HGF/c-Met: Dihexa binds to hepatocyte growth factor and amplifies its activity at the c-Met receptor.
- Promotion of synaptogenesis: Activation of this signaling pathway encourages the formation of new synaptic connections in the brain.
- Neurotrophic effects: The compound supports the survival and growth of neurons, similarly to neurotrophic factors such as BDNF (Brain-Derived Neurotrophic Factor).
Medical Applications (Research Status)
Dihexa is exclusively in the stage of preclinical and early clinical research. To date, there is no approved medical application for use in humans. In animal models, the following potential applications have been investigated:
- Cognitive impairment: Studies in Alzheimer models and age-related memory loss demonstrated improved learning and memory performance.
- Neurodegenerative diseases: Potential applications in Parkinson's disease and other neurodegenerative conditions are being explored.
- Heart failure: Due to the involvement of the HGF/c-Met pathway, cardioprotective effects have also been investigated.
Dosage and Administration
As Dihexa is not approved for human use, there are no official dosage recommendations. In animal studies, the substance has been administered orally, transdermally (through the skin), or by direct injection. In non-clinical communities (such as biohacker groups), transdermal applications are occasionally discussed; however, this is not medically recognized or recommended.
Side Effects and Safety Profile
The safety profile of Dihexa in humans is largely unknown, as comprehensive clinical trials are lacking. Potential risks include:
- Oncological risk: Since the HGF/c-Met signaling pathway is involved in cell proliferation, there are theoretical concerns regarding an increased tumor risk with long-term use.
