Dihexa is a synthetic, low-molecular-weight peptide derivative originally developed at Washington State University. It is derived from Angiotensin IV (Ang IV), a peptide fragment of the renin-angiotensin system. Dihexa was specifically engineered to readily cross the blood-brain barrier and exert its effects within the central nervous system. It belongs to the class of nootropic compounds — substances intended to support cognitive functions such as memory, learning, and mental performance.
The primary mechanism of action of Dihexa involves activation of the Hepatocyte Growth Factor receptor (HGF/c-Met) signaling pathway. Dihexa acts as a potentiating ligand that enhances HGF signaling, thereby stimulating synaptogenesis — the formation of new neuronal connections (synapses). This activity is believed to promote neural plasticity and thereby support learning and memory processes.
Dihexa is exclusively in the stage of preclinical and early clinical research. To date, there is no approved medical application for use in humans. In animal models, the following potential applications have been investigated:
