Drosha is an enzyme belonging to the RNase III family of ribonucleases, found in the nucleus of eukaryotic cells. It is a key component of the Microprocessor complex and is essential for the production of microRNAs (miRNAs) — small, non-coding RNA molecules that regulate gene expression. The human gene encoding this enzyme is officially designated DROSHA and is located on chromosome 5.
The biosynthesis of microRNAs begins in the cell nucleus. Here, primary microRNA (pri-miRNA) is first transcribed from DNA. Drosha, together with its cofactor DGCR8 (DiGeorge Syndrome Critical Region Gene 8), recognizes specific hairpin structures within the pri-miRNA and cleaves them precisely to generate the precursor miRNA (pre-miRNA).
Drosha therefore catalyzes the first and most critical processing step in canonical miRNA biogenesis.

Alterations in Drosha activity have been described in various cancers. Disrupted Drosha expression leads to impaired miRNA biogenesis, which can promote uncontrolled cell growth. Mutations in the DROSHA gene have been identified in nephroblastoma (Wilms tumor), certain ovarian tumors, and other solid tumors.
Since miRNAs play an important regulatory role in the nervous system, Drosha dysfunction has also been linked to the pathogenesis of neurological conditions, including certain neurodegenerative diseases.
Some viruses, including HIV and certain herpesviruses, interact with the Drosha Microprocessor complex in order to exploit or evade the cellular miRNA machinery for their own replication.
Drosha is considered a promising target in biomedical research. Approaches aimed at targeted modulation of Drosha activity could open up new therapeutic opportunities in the future, particularly in oncology and antiviral therapy. Furthermore, Drosha is being investigated as a potential biomarker for certain cancers, as its altered expression can be detected in tumor tissues.
