What is E280 (Propionic Acid)?

E280 is the official European food additive number for propionic acid (chemical formula: CH₃CH₂COOH). It is a short-chain fatty acid that occurs naturally in certain cheeses, fermented foods, and as a product of human metabolism. As a food additive, it is produced synthetically and is used primarily as a preservative to extend the shelf life of food products.

Origin and Natural Occurrence

Propionic acid occurs naturally in various foods, including Swiss-type cheeses such as Emmental, butter, and some fermented dairy products. It is also produced by intestinal bacteria in the human gut as part of normal fermentation processes. However, the propionic acid used in the food industry is predominantly manufactured through chemical synthesis or microbial fermentation.

Function and Mechanism of Action

E280 acts as an antimicrobial preservative. Its mechanism of action relies on the ability of the undissociated form of propionic acid to penetrate the cell membranes of moulds and certain bacteria. Once inside the microorganism, it lowers the intracellular pH and inhibits key metabolic enzymes, effectively preventing microbial growth and reproduction. Propionic acid is particularly effective against mould species of the genera Aspergillus and Penicillium.

Applications in Food

Under EU food additive regulation (EC) No. 1333/2008, E280 is approved for use in the following food categories:

  • Bread and bakery products (e.g., pre-packaged sliced bread, wrapped fine bakery goods)
  • Cheese and cheese products (for surface treatment)
  • Cereal-based products and certain breakfast cereals

Maximum permitted levels (MPL) vary by product category and are precisely defined in the EU regulation.

Related Additives (Propionates)

Propionic acid (E280) belongs to the propionates group of food additives, which also includes:

  • E281 – Sodium propionate
  • E282 – Calcium propionate
  • E283 – Potassium propionate

These salts of propionic acid are more commonly used in bakery products, as they are easier to incorporate into dough than the pure acid form.

Safety and Health Assessment

The European Food Safety Authority (EFSA) has evaluated propionic acid and its salts (E280–E283) and classified them as safe for human consumption at the approved levels. Since propionic acid is a natural metabolic product in the human body, it is normally broken down via fatty acid metabolism after ingestion. EFSA determined that a specific ADI (Acceptable Daily Intake) value was not necessary, as the substance is considered safe at typical dietary intake levels.

Possible Intolerances

In rare cases, sensitive individuals have reported headaches or migraines following consumption of foods containing propionic acid. However, scientific evidence for a clinically relevant intolerance to E280 at food-level doses remains limited. Individuals who suspect a sensitivity should monitor their intake accordingly.

Labelling Requirements

In the EU, E280 must be declared on food ingredient lists either by its E-number (E280) or by the functional class name followed by the specific name (preservative: propionic acid).
