E421 is the European approval number for mannitol, a naturally occurring sugar alcohol (polyol). Mannitol is found in a variety of plants, algae, and fungi, including ash tree exudate, celery, mushrooms, and olives. For industrial purposes, it is primarily produced by the hydrogenation of fructose or through microbial fermentation.

Mannitol provides approximately 1.6 kcal per gram, significantly less than regular sugar (4 kcal/g). Because it is poorly absorbed in the small intestine, it has a low glycemic index and has minimal impact on blood sugar levels. This makes it a popular alternative for individuals with diabetes mellitus. The unabsorbed portion passes into the large intestine, where it is partially fermented by gut bacteria.

The European Food Safety Authority (EFSA) has classified E421 as safe for use in food. No Acceptable Daily Intake (ADI) value has been established, as mannitol occurs naturally in foods and no toxicological concerns have been identified. Its use is regulated in the EU under Regulation (EC) No 1333/2008.
