E629 is the EU authorization number for potassium 5´-inosinate (also known as dipotassium inosinate), a nucleotide-based flavor enhancer. It belongs to the group of 5´-ribonucleotides and is used in the food industry to intensify the so-called umami taste — widely recognized as the fifth basic taste alongside sweet, sour, salty, and bitter.

Origin and Production

Potassium 5´-inosinate can be derived from animal sources such as fish and meat, or produced biotechnologically through fermentation. It is closely related to other nucleotide flavor enhancers, including E627 (disodium guanylate) and E631 (disodium inosinate). In practice, these compounds are frequently used in combination with E621 (monosodium glutamate), as they act synergistically and can significantly amplify the overall flavor-enhancing effect.

Use in Food Products

E629 is typically found in the following types of products:

  • Ready-made meals and frozen foods
  • Instant soups and stock cubes
  • Snacks, chips, and savory crackers
  • Sauces, dressings, and seasoning blends

Mechanism of Action

Potassium 5´-inosinate works by binding to umami receptors (particularly TAS1R1/TAS1R3 receptors) on the tongue. The nucleotides enhance the perception of glutamates and other flavor compounds without having a pronounced taste of their own. When combined with glutamates, the flavor intensity can be multiplied many times over.

Safety and Tolerability

E629 has been evaluated by the European Food Safety Authority (EFSA) and the Joint FAO/WHO Expert Committee on Food Additives (JECFA). No specific Acceptable Daily Intake (ADI) value was established, as the substance was considered safe under normal conditions of use. However, certain groups should exercise caution:

  • Gout patients: Inosinate is a purine derivative and may increase uric acid levels in the blood, potentially triggering gout attacks.
  • Individuals sensitive to glutamates: Since E629 is often used alongside glutamates, sensitive individuals may report symptoms such as headaches or nausea, although a clear causal link has not been scientifically confirmed.
  • People with asthma: Reactions in asthma patients have been reported in rare cases.

Labeling Requirements

Under EU food law, E629 must be declared in the ingredient list of food products, either as E629 or as potassium 5´-inosinate. Consumers who wish to avoid nucleotide flavor enhancers should read the ingredient lists of processed foods carefully.
