What are follicular dendritic cells?

Follicular dendritic cells (FDCs) are highly specialized, non-hematopoietic stromal cells of lymphoid tissue. They are found primarily in the germinal centers of secondary lymphoid organs such as lymph nodes, the spleen, and tonsils. Despite sharing the term "dendritic" with classical dendritic cells, FDCs belong to an entirely different cell lineage and serve distinct immunological functions.

Structure and characteristics

Follicular dendritic cells are characterized by their long, branching projections (dendrites) that form a dense network within germinal centers. This network allows them to retain and display antigens on their surface for extended periods. Unlike classical dendritic cells, FDCs do not express significant levels of MHC class II molecules and are therefore not directly involved in T cell activation. Instead, they bind antigen-antibody complexes and antigen-complement complexes via specific surface receptors.

  • Fc receptors: Bind antigen-antibody complexes (immune complexes)
  • Complement receptors (CR1, CR2): Bind complement-opsonized antigens
  • Long-term antigen retention: FDCs can display antigens on their surface for weeks to months

Function in the immune system

The primary role of follicular dendritic cells is to support B cell maturation within germinal centers. After initial activation by an antigen, B cells migrate into germinal centers, where they interact intensively with FDCs. This process involves several critical steps:

  • Antigen presentation: FDCs present antigens as immune complexes on their surface, enabling the selection of B cells that produce antibodies with increasingly high binding affinity.
  • Affinity maturation: B cells that successfully recognize antigen on FDCs are positively selected and survive, while those with low affinity undergo apoptosis.
  • Memory B cell formation: FDCs contribute to the generation of memory B cells, which enable a rapid immune response upon re-exposure to the same antigen.
  • Plasma cell differentiation: Through interaction with FDCs, B cells mature into antibody-secreting plasma cells.

Importance for the immune response

Follicular dendritic cells are essential for the humoral immune response -- the arm of the immune system based on the production of antibodies. Without functional FDCs, germinal center reactions, affinity maturation, and immunological memory would be severely impaired, making FDCs an indispensable component of the adaptive immune system.

Clinical relevance

Dysfunction or transformation of follicular dendritic cells has been associated with several diseases:

  • Follicular dendritic cell sarcoma: A rare malignant tumor arising from FDCs, typically in lymph nodes or extranodal sites. Symptoms include enlarged lymph nodes and systemic signs of illness.
  • HIV infection: HIV can persist bound to FDCs within germinal centers, using them as a long-lived reservoir for further viral spread.
  • Autoimmune diseases: Aberrant germinal center activity involving FDCs may contribute to the generation of autoreactive antibodies.

Distinction from classical dendritic cells

Although both cell types share the term "dendritic," they differ fundamentally:

  • Classical dendritic cells originate from hematopoietic precursors and primarily activate T cells.
  • Follicular dendritic cells are of mesenchymal origin and primarily interact with B cells.
  • FDCs do not express meaningful levels of MHC class II molecules and do not take up antigens by phagocytosis.
