Glycan targeting is an innovative strategy in medical research and therapy in which specific sugar structures (glycans) on cell surfaces are used as target structures for active substances, antibodies or diagnostic markers.
Glycans are complex carbohydrates that are covalently bound to proteins or lipids and play a crucial role in cell-cell communication, immune response, inflammation and tumour progression. Especially in cancer research and immunotherapy, the targeted targeting of glycan structures is becoming increasingly important. In glycan targeting, abnormal glycosylation patterns, which often occur on the surface of tumour cells, infected or chronically inflamed cells, are recognised and therapeutically addressed. These altered sugar structures - such as sialic acids, Lewis antigens or O-glycosylated mucins - can act as tumour-associated antigens (TAA).
Therapeutic approaches include the development of monoclonal antibodies, glycan-binding peptides, nanoparticles or CAR-T cells that are customised to these structures. The aim is to selectively recognise and eliminate pathological cells while protecting healthy tissue. Glycan targeting is also increasingly being used in the field of vaccinology and infection biology - e.g. for the targeted activation of immune cells, for improving antigen uptake or for blocking pathogenic interactions between bacteria/viruses and human cells. The greatest challenge lies in the high structural diversity and dynamic nature of glycan complexes, which makes precise target definition difficult. Nevertheless, glycan targeting opens up promising perspectives for personalised medicine, cancer treatment and immunomodulation.
