What is Heneicosapentaenoic Acid?

Heneicosapentaenoic acid (abbreviated HPA) is a polyunsaturated fatty acid belonging to the omega-3 fatty acid family. Its systematic name is 21:5(n-3), indicating a carbon chain of 21 carbon atoms with five double bonds. It is among the rarer members of the long-chain omega-3 fatty acids and differs structurally from the more widely known representatives eicosapentaenoic acid (EPA, 20:5) and docosahexaenoic acid (DHA, 22:6).

Occurrence and Sources

Heneicosapentaenoic acid is found primarily in marine organisms. Relevant natural sources include:

  • Marine algae and phytoplankton, particularly certain diatom species
  • Marine invertebrates such as mollusks and crustaceans
  • In small amounts in certain fish species
  • Certain cyanobacteria (blue-green algae)

The concentration of HPA in most foods is very low compared to EPA and DHA, meaning that significant amounts are rarely consumed through a regular diet.

Biochemistry and Mechanism of Action

Like other omega-3 fatty acids, heneicosapentaenoic acid is believed to act through several biochemical pathways:

  • Incorporation into cell membranes: HPA can be incorporated into membrane phospholipids, influencing their fluidity and signal transduction.
  • Lipid mediator formation: Omega-3 fatty acids serve as precursors for bioactive lipid mediators that can support anti-inflammatory processes.
  • Competition with arachidonic acid: Like EPA, HPA may structurally compete with the pro-inflammatory arachidonic acid, potentially promoting a more balanced ratio of inflammatory mediators.
  • Gene expression: Via receptors such as PPAR-alpha (peroxisome proliferator-activated receptor alpha), long-chain omega-3 fatty acids can regulate lipid metabolism and inflammatory responses.

Potential Health Significance

Research into heneicosapentaenoic acid is still at an early stage. Initial scientific investigations suggest the following possible properties:

  • Anti-inflammatory effects: Like other omega-3 fatty acids, HPA may possess anti-inflammatory properties.
  • Cardiovascular health: Long-chain omega-3 fatty acids are generally associated with supporting heart health.
  • Neuroprotective effects: Due to its structural similarity to DHA, HPA is being studied for neuroprotective properties.
  • Antioxidant activity: Some studies suggest a potential antioxidant effect.

Important note: Most of these findings come from in vitro studies or animal experiments. Clinical studies in humans specifically examining HPA remain limited. Therefore, no definitive recommendations for HPA supplementation can be made at this time.

Distinction from EPA and DHA

Heneicosapentaenoic acid is often discussed in the context of the well-researched omega-3 fatty acids EPA (eicosapentaenoic acid) and DHA (docosahexaenoic acid). In contrast to these:

  • HPA has a carbon chain one unit longer than EPA (21 vs. 20 carbon atoms).
  • The scientific evidence base for HPA is considerably thinner than for EPA and DHA.
  • There are currently no official dietary intake recommendations for HPA from bodies such as the WHO or EFSA.

Safety and Toxicity

Toxicological data specifically relating to heneicosapentaenoic acid in humans are largely unavailable. Based on the general safety profile of omega-3 fatty acids, HPA is considered safe in usual dietary amounts. When taking supplements containing concentrated omega-3 fatty acids, caution is always advisable, and consultation with a qualified healthcare professional is recommended.
