IL-10, also known as Interleukin-10, is an endogenous cytokine – a signaling molecule of the immune system. It is produced by various immune cells, including macrophages, T cells, B cells, and dendritic cells. IL-10 is considered one of the most important anti-inflammatory cytokines in the human body and plays a central role in controlling inflammatory responses.

An imbalance in IL-10 production is associated with various conditions. A deficiency of IL-10 or reduced cellular responsiveness to IL-10 can result in excessive inflammation and has been observed in conditions such as inflammatory bowel disease (e.g., Crohn's disease, ulcerative colitis), rheumatoid arthritis, and other autoimmune disorders.
During infections, IL-10 regulates the balance between an effective immune response and the prevention of tissue damage. However, excessive IL-10 production can impair the clearance of pathogens – a phenomenon described in chronic infections such as tuberculosis and malaria.
In cancer research, IL-10 plays an ambivalent role. On one hand, it can inhibit tumor growth; on the other, tumor cells may exploit IL-10 to evade immune recognition.
Due to its anti-inflammatory properties, IL-10 is being investigated as a potential therapeutic agent for various conditions. Recombinant IL-10 (ilodecakin) has been studied in clinical trials involving patients with inflammatory bowel disease and other conditions. Additionally, IL-10 analogues and substances that promote IL-10 production are being explored as therapeutic targets. Conversely, targeted inhibition of IL-10 may be beneficial in cancer immunotherapy to enhance immune responses against tumor cells.
