The immune system comprises various cell types, including T cells, B cells, natural killer (NK) cells, and macrophages. Over time, these cells accumulate damage, lose their ability to divide, and so-called senescent cells – cells that have stopped functioning properly but remain alive – begin to accumulate. These senescent cells release pro-inflammatory signaling molecules, contributing to a state of chronic low-grade inflammation known as inflammaging.

Studies suggest that moderate caloric restriction and intermittent fasting can stimulate autophagy – the process by which damaged cellular components are broken down and recycled. This cellular housekeeping mechanism may contribute to the renewal and optimization of immune cells.

Exercise and Physical Activity

Regular, moderate physical activity has well-documented positive effects on the immune system. It promotes the circulation of immune cells, reduces inflammatory markers, and may help preserve telomere length – a key indicator of cellular health and longevity.

Senolysis and Senolytic Agents

A promising scientific approach involves the targeted elimination of senescent cells using compounds called senolytics. Substances such as quercetin, dasatinib, and navitoclax are being investigated in preclinical and early clinical studies to improve immune function by clearing dysfunctional cells.

Thymus Peptides and Thymic Regeneration

Since the thymus – the primary organ for T cell maturation – shrinks with age (thymic involution), some research efforts focus on regenerating this tissue. The TRIIM pilot study explored a combination of growth hormone, DHEA, and metformin and found early evidence of thymic tissue rejuvenation.

Stem Cell Therapies

Experimental approaches using hematopoietic (blood-forming) stem cells aim to fundamentally renew the immune system. However, these methods remain in early stages of development and are not yet available for broad clinical use.
