What is Melanotan II?

Melanotan II (also known as MT-II or MT2) is a synthetically produced peptide structurally similar to the naturally occurring hormone alpha-melanocyte-stimulating hormone (α-MSH). It was originally developed in the 1980s at the University of Arizona as a research compound intended to reduce skin cancer risk from UV radiation. Melanotan II is not approved as a medicinal product in the European Union, the United States, or most other countries, and is considered an illegal lifestyle substance.

Mechanism of Action

Melanotan II binds to melanocortin receptors in the body, particularly receptor subtypes MC1R, MC3R, MC4R, and MC5R. By activating the MC1R receptor in melanocytes (the pigment-producing cells of the skin), it stimulates the production of melanin – the pigment responsible for skin color and UV protection. This results in skin darkening, sometimes even without direct sun exposure.

Through the MC4R receptor, Melanotan II also affects the central nervous system, causing side effects such as appetite suppression, nausea, and – one of its commonly advertised effects – increased sexual arousal. This latter effect led to the development of a related compound called bremelanotide (PT-141), which is approved in the United States for treating sexual dysfunction.

Use and Prevalence

Melanotan II is typically sold as a powder that users dissolve in water and self-administer via subcutaneous injection. It is also available as a nasal spray, although its bioavailability via this route is considered poor. It is commonly used in the bodybuilding community and among individuals seeking rapid skin tanning.

Health Risks and Side Effects

Since Melanotan II is not clinically approved, comprehensive long-term safety data are lacking. Known and potential side effects include:

  • Spontaneous erections in men
  • Changes to moles (naevus changes) – potentially carcinogenic
  • Blood pressure changes and cardiovascular risks
  • Headaches and dizziness
  • Infection risks from improper self-injection (e.g., HIV, hepatitis)
  • Psychological dependence and uncontrolled use

Of particular concern is the possibility that Melanotan II may alter existing moles or promote the growth of melanocytic lesions, potentially increasing the risk of malignant melanoma (skin cancer).

Legal Status

In the European Union and most other countries, Melanotan II is not approved as a medicinal product. Its sale and distribution as a medicine are illegal. Nevertheless, the substance is widely available through unregulated online platforms and grey markets. The European Medicines Agency (EMA) and national regulatory authorities explicitly warn against its use.
