Proteolysis is the enzymatic breakdown of proteins into smaller peptides or amino acids.
It occurs both inside cells and extracellularly, playing a crucial role in protein metabolism, inflammation control, and cellular turnover. Without proteolysis, the body could not remove damaged or excess proteins.
Key enzymes involved are proteases and peptidases, which cleave protein chains. Proteolysis occurs via several systems—most notably the proteasome and lysosomal pathways (as part of autophagy). These mechanisms help regulate the immune response, control cell proliferation, and eliminate dysfunctional proteins.
In digestion, enzymes like pepsin and trypsin initiate proteolysis in the gastrointestinal tract. Dysregulated proteolysis is linked to diseases such as cancer, muscle wasting, and autoimmune disorders.
