Pterostilbene is a natural plant compound from the stilbene family, found primarily in blueberries and grapes. It is a bioactive polyphenol known for its antioxidant and anti-inflammatory properties.
Pterostilbene is a naturally occurring polyphenol belonging to the stilbene chemical class. It is structurally closely related to the well-known compound resveratrol, but differs by having two methoxy groups instead of hydroxyl groups. This structural difference gives pterostilbene greater bioavailability and lipophilicity. Pterostilbene was first identified in sandalwood and is found in particularly high concentrations in blueberries, cranberries, grapes, and peanuts.
Pterostilbene exerts its biological effects through several molecular pathways:
Studies suggest that pterostilbene may have positive effects on blood pressure and lipid profiles. It has been shown to help lower LDL cholesterol levels and improve HDL levels. It also provides vascular protective effects by inhibiting lipid oxidation and inflammatory processes in blood vessel walls.
Preclinical and clinical studies have demonstrated blood sugar-lowering effects of pterostilbene. It improves insulin sensitivity and inhibits enzymes involved in carbohydrate digestion, making it a promising area of research for type 2 diabetes prevention.
Pterostilbene has shown neuroprotective effects in animal models. It may improve cognitive function, reduce oxidative stress in the brain, and slow neurodegenerative processes. Researchers are currently investigating its potential role in age-related cognitive decline and neurodegenerative diseases such as Alzheimer disease.
Laboratory studies indicate that pterostilbene can inhibit the growth of various cancer cell lines, including breast, colon, and pancreatic cancer. The mechanisms include promoting apoptosis (programmed cell death), inhibiting cell division, and anti-angiogenic effects. However, these findings are primarily from cell culture and animal studies; clinical trials in humans are still largely pending.
Through activation of the AMPK signaling pathway, pterostilbene may positively influence fat metabolism and contribute to body weight regulation. Positive effects on fat storage and energy expenditure have been observed in animal models.
A key advantage of pterostilbene over resveratrol is its significantly higher bioavailability. Resveratrol is rapidly metabolized and conjugated in the body, limiting its effectiveness. Pterostilbene, being more lipophilic (fat-soluble), is better absorbed from the intestine and broken down more slowly. Studies estimate the bioavailability of pterostilbene at approximately 80 percent, compared to less than 1 percent for unchanged resveratrol.
Pterostilbene is available as a dietary supplement in capsule or tablet form. Clinical studies have used dosages ranging from 50 mg to 250 mg per day, with lower doses (50 mg/day) commonly used for general antioxidant purposes. Higher doses have been studied for blood sugar and blood pressure regulation. It is recommended to take pterostilbene with a fat-containing meal to enhance absorption.
Pterostilbene is generally considered well tolerated at moderate doses. Some studies have noted slight increases in LDL cholesterol at higher dosages, which remains a topic of ongoing research. Other possible side effects at very high doses include:
Pregnant women, breastfeeding mothers, and individuals taking medications should consult a physician before taking pterostilbene supplements.
Pterostilbene is found in various foods, though in relatively small amounts:
The amount of pterostilbene obtained through diet is generally much lower than the therapeutic doses used in studies, which is why targeted supplementation may be considered for specific health goals.
