What is Sermorelin?

Sermorelin is a synthetically produced peptide that closely resembles the naturally occurring growth hormone-releasing hormone (GHRH) in the human body. It consists of the first 29 amino acids of endogenous GHRH and stimulates the pituitary gland to increase the secretion of growth hormone (GH). Sermorelin belongs to the class of GHRH analogues and is used for both diagnostic and therapeutic purposes.

Mechanism of Action

Sermorelin binds to specific GHRH receptors in the pituitary gland, activating somatotropic cells that are responsible for the production and release of growth hormone. Unlike direct administration of recombinant growth hormone, Sermorelin stimulates the body's own growth hormone production in a physiological manner. This mechanism largely preserves the natural feedback regulation of the body, reducing the risk of excess growth hormone production.

Medical Indications

Diagnosis of Growth Hormone Deficiency

Sermorelin is used as a stimulation test to assess pituitary function. After injection of Sermorelin, growth hormone levels in the blood are measured to confirm or rule out a deficiency.

Treatment of Growth Hormone Deficiency in Children

In children with confirmed growth hormone deficiency, Sermorelin can be used to promote linear growth by stimulating the pituitary gland to produce more of the body's own growth hormone.

Use in Adults

In adults, Sermorelin is studied and used in some countries to address the age-related decline in growth hormone production, as well as to support body composition and metabolism. Regulatory approval status varies by country.

Dosage and Administration

Sermorelin is typically administered as a subcutaneous injection (under the skin), often in the evening before bedtime, as natural growth hormone secretion is highest during sleep. The exact dosage depends on body weight, age, and therapeutic goal, and is determined individually by the treating physician. Typical doses range from 0.2 to 0.3 mg per day, but may vary depending on the clinical situation.

Side Effects

Sermorelin is generally well tolerated. Possible side effects include:

  • Local reactions at the injection site (redness, swelling, pain)
  • Flushing (feeling of warmth, skin redness)
  • Headache
  • Nausea
  • Difficulty swallowing (rare)
  • Dizziness

Serious side effects are rare. Sermorelin should not be used in individuals with known hypersensitivity to the substance or any of its components.

Contraindications and Interactions

Sermorelin should not be used in the following situations:

  • Known hypersensitivity to the active ingredient
  • Active tumors or certain cancers (as growth hormone may promote tumor growth)
  • Pregnancy and breastfeeding (insufficient data available)

Potential interactions exist with glucocorticoids, thyroid hormones, insulin, and other hormones that may influence growth hormone secretion. Careful medical monitoring is required during treatment.
