The Simian Immunodeficiency Virus (SIV) is a retrovirus that naturally infects various non-human primates, including chimpanzees, gorillas, mandrills, and African green monkeys. SIV is widely regarded as the evolutionary origin of the Human Immunodeficiency Virus (HIV), the virus responsible for AIDS in humans. Studying SIV has provided critical insights into the origins, transmission, and treatment of HIV/AIDS.
Scientists believe that HIV emerged through zoonotic transmission – the transfer of a pathogen from animals to humans. It is thought that people in Central Africa came into contact with SIV-infected primates through hunting or the handling of bushmeat. Over time, the virus adapted to the human host and evolved into HIV.
SIV belongs to the family Retroviridae and the genus Lentivirus. Like HIV, SIV primarily targets CD4-positive T lymphocytes – a subset of white blood cells that are essential for immune defense. The virus integrates its RNA into the host cell DNA using the enzyme reverse transcriptase, allowing it to replicate within the host.
Notably, many natural hosts of SIV – such as African green monkeys – do not develop an AIDS-like illness despite being infected. This is due to an evolutionary adaptation in which the immune system suppresses the chronic inflammatory response that leads to immune failure in HIV-infected humans.
SIV is an indispensable model in biomedical research. Studies using SIV-infected primates allow scientists to better understand HIV pathogenesis and to develop new therapies and vaccine candidates.
Although SIV is structurally and genetically closely related to HIV, direct transmission of SIV to humans is extremely rare and has been poorly documented. The virus is primarily adapted to non-human primates. However, isolated case reports describe laboratory workers or veterinarians testing seropositive for SIV antibodies after close contact with infected primate material, without developing a stable infection or disease.
