Tachyphylaxis refers to the rapid loss of efficacy of a drug after repeated use, typically within hours to days.
Unlike gradual drug tolerance, tachyphylaxis develops suddenly and is often seen with nasal decongestants, antidepressants (SSRIs), beta-blockers, and hormonal agents.
The underlying cause is usually receptor desensitization or downregulation—meaning fewer receptors are available or responsive. As a result, patients may no longer experience therapeutic benefits despite consistent dosage. For example, nasal sprays can lead to rebound congestion due to overuse and tachyphylaxis.
Preventing tachyphylaxis involves treatment breaks, drug rotation, or switching to non-pharmacological options. In the case of SSRIs, tachyphylaxis may indicate adaptive changes in CNS receptors and require therapeutic reassessment.
Medical Implications of Tachyphylaxis
- Sudden loss of drug effect despite unchanged dosing
- Common with nasal sprays, antidepressants, hormones, beta-blockers
- Caused by receptor desensitization or reduced receptor availability
- Avoidable through intermittent use or switching medications
- May signal need for clinical reevaluation in psychiatric or endocrine therapy
