A tissue growth factor is a naturally occurring signaling protein that regulates cell growth, proliferation, differentiation, and survival within tissues. These proteins act as biological messengers, binding to specific receptors on cell surfaces and triggering intracellular signaling cascades that control how cells behave and respond to their environment.

Growth factors are present in virtually all tissues of the human body and are essential for normal development, tissue repair, and the maintenance of tissue homeostasis. Well-known examples include the Epidermal Growth Factor (EGF), the Transforming Growth Factor (TGF), the Vascular Endothelial Growth Factor (VEGF), and the Fibroblast Growth Factor (FGF).

Biological Functions

Tissue growth factors serve a wide range of functions in the human body:

  • Cell proliferation: Stimulation of cell division and growth in damaged or renewing tissues.
  • Cell differentiation: Guiding stem cells to specialize into specific tissue types.

Epidermal Growth Factor (EGF)

The Epidermal Growth Factor promotes the proliferation of skin, mucous membrane, and glandular cells. It is particularly important for the regeneration of skin and mucous membranes and plays a key role in wound healing. EGF is actively used in modern dermatology and cosmetic medicine.

Transforming Growth Factor (TGF)

The Transforming Growth Factor exists in two main forms: TGF-alpha and TGF-beta. TGF-beta plays a particularly significant role in regulating inflammatory processes, scar formation, and immune suppression. Overactivation of TGF-beta is associated with fibrotic diseases.

Vascular Endothelial Growth Factor (VEGF)

The Vascular Endothelial Growth Factor is the most important regulator of angiogenesis. It stimulates the growth of new blood vessels, which plays a critical role in both wound healing and tumor development. Anti-VEGF therapies are therefore used in oncology and ophthalmology.

Fibroblast Growth Factor (FGF)

Clinical Significance

Tissue growth factors are not only physiologically important but also highly relevant in clinical medicine. Disruptions in their regulation can lead to a range of diseases:

  • Cancer: Many types of cancer are associated with overexpression or mutation of growth factor receptors (e.g., HER2 in breast cancer, EGFR in lung carcinoma).
  • Fibrosis: Excessive TGF-beta activity can lead to pathological overgrowth of connective tissue in organs such as the lungs, liver, or kidneys.
  • Chronic wound healing disorders: A deficiency of growth factors can impair the healing of chronic wounds such as diabetic foot syndrome.
  • Eye diseases: Overproduction of VEGF causes pathological neovascularization in wet macular degeneration and diabetic retinopathy.

Therapeutic Applications

Knowledge of tissue growth factors has led to the development of innovative therapies:

  • Growth factor-based wound therapy: Topical application of EGF or PDGF (platelet-derived growth factor) for the treatment of chronic wounds.
  • Anti-VEGF therapy: Drugs such as bevacizumab or ranibizumab block VEGF and are used in cancer and eye diseases.
  • Tyrosine kinase inhibitors: These inhibit growth factor receptors and are used as targeted cancer therapies (e.g., imatinib, erlotinib).
  • Regenerative medicine: Growth factors are used in tissue engineering research to promote tissue replacement and organ regeneration.

Diagnosis and Laboratory Testing

Tissue growth factors can be measured in blood, tissue, or body fluids using laboratory diagnostic methods. Elevated or reduced levels may indicate certain diseases. In tumor diagnostics, growth factor receptors such as HER2 or EGFR serve as important biomarkers that influence the choice of therapy.
