What is Periodic Acid?

Periodic acid (chemical formula: HIO4 or H5IO6 in its orthoperiodic form) is a strong inorganic oxidizing acid in which iodine is present in its highest oxidation state (+7). It exists in two main forms: metaperiodic acid (HIO4) and orthoperiodic acid (H5IO6). In medicine and the life sciences, it is best known as a key reagent in histology and biochemistry.

Chemical Properties

Periodic acid is a colorless to white crystalline solid that dissolves readily in water. As a strong oxidizing agent, it reacts selectively with vicinal diols (adjacent hydroxyl groups), which are commonly found in sugar molecules and polysaccharides. This reaction, known as the Malaprade reaction or periodate cleavage, cleaves the carbon-carbon bond between adjacent hydroxyl groups to form aldehyde groups.

Medical and Biochemical Applications

PAS Staining in Histology

The most significant medical application of periodic acid is the Periodic Acid-Schiff (PAS) staining reaction. In this histochemical method, periodic acid oxidizes polysaccharides and glycoproteins in tissue sections, generating aldehyde groups. These then react with Schiff reagent to produce a characteristic magenta-red coloration. PAS staining is widely used in clinical pathology and research to visualize:

  • Glycogen in liver and muscle cells
  • Basement membranes in renal pathology
  • Mucus substances (mucins) in glandular cells
  • Fungi and other microorganisms with polysaccharide-rich cell walls
  • Glycoprotein complexes in various tissues

Biochemical Analysis

In biochemical research, periodic acid is used for the selective oxidation of carbohydrates, enabling structural analysis of sugar chains, labeling of glycoproteins, and quantification of polysaccharides.

Molecular Biology and Proteomics

Periodic acid is also applied for the modification of glycoproteins, for example in conjugating antibodies with enzymes or fluorescent labels in immunohistochemistry.

Safety Information

Periodic acid is a strong oxidizing agent and must be handled with care. The following safety precautions apply in laboratory settings:

  • Avoid contact with skin, eyes, and mucous membranes
  • Wear protective gloves, safety goggles, and a lab coat
  • Keep away from flammable substances
  • Store in a cool, dry location, separated from organic materials
  • In case of skin contact, rinse immediately with plenty of water

Periodic acid is not a pharmaceutical drug and is not applied directly to patients. It is used exclusively as a laboratory and research reagent.

Clinical Relevance

Through its role in PAS staining, periodic acid indirectly contributes to important clinical diagnostics. In nephrology, PAS staining is used to evaluate kidney biopsies and diagnose conditions such as diabetic nephropathy, IgA nephritis, and other glomerular diseases. In infectious disease pathology, it aids in identifying fungal infections in tissue samples.
