What is Kisspeptin-10?

Kisspeptin-10 (also known as KP-10) is the shortest and biologically highly active form of the kisspeptin family, a group of neuropeptides encoded by the KISS1 gene. It consists of 10 amino acids and represents the C-terminal fragment of the longer precursor protein kisspeptin-54. Kisspeptin-10 binds to the G protein-coupled receptor KISS1R (also known as GPR54) and is an essential regulator of the reproductive axis in the human body.

Mechanism of Action

Kisspeptin-10 acts primarily on specialized nerve cells in the hypothalamus known as GnRH neurons (gonadotropin-releasing hormone neurons). By binding to KISS1R, kisspeptin-10 stimulates the release of GnRH, which in turn prompts the pituitary gland to secrete LH (luteinizing hormone) and FSH (follicle-stimulating hormone). These hormones regulate gonadal function and thereby the production of sex hormones such as testosterone and estrogen.

Reproductive Medicine

Kisspeptin-10 plays a key role in puberty, the menstrual cycle, ovulation, and male fertility. Disruptions in the kisspeptin system can lead to hypogonadotropic hypogonadism, a condition in which the gonads are underactive due to insufficient stimulation by the brain and pituitary gland. In women, a deficiency in this system can result in absent ovulation and infertility.

Diagnostic Use

In clinical research, kisspeptin-10 is used as a diagnostic tool to assess the responsiveness of the hypothalamic-pituitary-gonadal (HPG) axis. An intravenous or subcutaneous dose of kisspeptin-10 can stimulate an LH pulse, providing insights into the functional integrity of the reproductive system.

Therapeutic Potential

Clinical studies are investigating the use of kisspeptin-10 as a therapeutic agent for:

  • Female infertility (e.g., triggering ovulation in in vitro fertilization)
  • Hypogonadism in men and women
  • Hypothalamic amenorrhea
  • Delayed puberty

Of particular interest is the use of kisspeptin-10 as an alternative to human chorionic gonadotropin (hCG) for triggering ovulation during IVF treatment, as it may reduce the risk of ovarian hyperstimulation syndrome (OHSS).

Occurrence and Regulation

Kisspeptin-10 is produced endogenously in the hypothalamus and the placenta. During pregnancy, kisspeptin levels in the blood rise significantly, suggesting a role in the maintenance of pregnancy. Secretion is regulated by sex hormones (positive and negative feedback), nutritional status, stress, and circadian rhythms.

Side Effects and Safety

Kisspeptin-10 is generally well tolerated in clinical studies. Reported side effects are rare and include mild local reactions at the injection site and transient changes in blood pressure. Because the peptide is broken down very rapidly (short half-life of a few minutes), systemic long-term side effects have not been observed to date. Long-term safety studies on repeated administration are still ongoing.
