What is MGF?
MGF (Mechano Growth Factor) is an isoform of Insulin-like Growth Factor 1 (IGF-1). It is produced through alternative splicing of the IGF-1 messenger RNA (mRNA) and is primarily expressed in mechanically loaded muscle, bone, and cardiac tissue. MGF plays a central role in the regeneration and growth of skeletal muscle cells.
Mechanism of Action
When muscle tissue is subjected to mechanical stress through physical exercise, stretching, or micro-injuries, MGF is released locally in the affected tissue. The growth factor binds to specific receptors on muscle precursor cells known as satellite cells, activating them and triggering the following processes:
- Proliferation (multiplication) of satellite cells
- Differentiation into mature muscle cells (myocytes)
- Integration of new cell nuclei into existing muscle fibres (muscle hypertrophy)
- Inhibition of programmed cell death (apoptosis) in muscle cells
MGF acts primarily in a local (paracrine and autocrine) manner, meaning it mainly influences cells in the immediate vicinity of its site of production.
Biological Significance
MGF is particularly important for:
- Muscle regeneration following injuries or intense training
- Muscle preservation with ageing (prevention of sarcopenia)
- The response of muscle tissue to strength and endurance training
- Regeneration of cardiac muscle tissue after a myocardial infarction (experimental)
As the body ages, MGF secretion decreases, which partly explains the age-related loss of muscle mass known as sarcopenia.
MGF in Sport and Research
Due to its muscle-building properties, MGF is a focus of sports medicine and pharmaceutical research. Synthetically produced MGF peptides (e.g., PEGylated MGF) are being studied in preclinical trials for their therapeutic potential, including treatment of muscular dystrophy, age-related muscle wasting, and support of muscle recovery after surgery.
In competitive sports, MGF is considered a potential doping agent. The World Anti-Doping Agency (WADA) lists MGF and its analogues on the prohibited list, as misuse can lead to undesirable increases in muscle mass and athletic performance.
Medical Potential and Clinical Research
Medical research is investigating the therapeutic use of MGF in the following areas:
- Muscular dystrophies: Genetically caused muscle diseases in which muscle regeneration is impaired
- Sarcopenia: Age-related decline in muscle mass and strength
- Cardiac regeneration: Supporting healing after myocardial infarction

Safety and Risks
The uncontrolled or abusive use of synthetic MGF peptides without medical supervision carries significant risks, as long-term safety data in humans are largely absent. Potential risks include:
