What is Allantoin?

Allantoin (chemical name: 5-ureidohydantoin) is a nitrogen-containing organic compound found naturally in several plants – most notably in comfrey (Symphytum officinale) – as well as in the urine of many mammals. The human body produces only negligible amounts of allantoin on its own. In pharmaceutical and cosmetic manufacturing, allantoin is obtained either synthetically or from plant-based sources and is used across a wide range of applications.

Mechanism of Action

Allantoin exerts its effects through several biological mechanisms:

  • Keratolytic effect: Allantoin softens and loosens the bonds between dead skin cells, resulting in smoother, more supple skin.
  • Cell proliferation: The compound stimulates cell division and tissue growth, supporting wound healing and skin regeneration.
  • Moisture retention: Allantoin acts as a humectant, improving skin hydration by attracting and retaining water.
  • Soothing effect: It reduces skin irritation and inflammatory responses, making it especially suitable for sensitive skin.

Natural Sources

Allantoin occurs naturally in:

  • Comfrey (Symphytum officinale) – particularly abundant in the roots and leaves
  • Sugar beet (Beta vulgaris)
  • Chamomile (Matricaria chamomilla)
  • Wheat germ
  • Urine of many mammals (e.g., cattle, horses)

Medical and Cosmetic Applications

Dermatology and Wound Care

In dermatology, allantoin is incorporated into creams, ointments, and gels to accelerate wound healing, improve scar tissue appearance, and calm irritated or inflamed skin. It is particularly useful in treating:

  • Dry and rough skin conditions
  • Minor burns and sunburn
  • Psoriasis and eczema
  • Abrasions and minor wounds
  • Post-surgical scar management

Cosmetics

Allantoin is a widely used ingredient in body lotions, facial creams, lip care products, deodorants, shampoos, and after-sun preparations. It improves skin texture, reduces redness, and is featured in anti-aging products due to its ability to promote skin cell renewal.

Oral and Systemic Use

In some countries, allantoin is also used orally as a dietary supplement or incorporated into mouthwashes, where it has demonstrated anti-inflammatory properties in oral tissues. However, research into systemic applications remains limited.

Recommended Dosage and Concentration

In topical formulations (creams and ointments), allantoin is typically used at concentrations of 0.1% to 2%. Higher concentrations may be found in specialized wound care preparations. In cosmetic products, typical concentrations range from 0.1% to 0.5%.

Safety and Tolerability

Allantoin is considered extremely well-tolerated and safe. It is recognized by the U.S. Food and Drug Administration (FDA) as a safe active ingredient in over-the-counter (OTC) skin protectant products and is listed as a permitted cosmetic ingredient by the European Commission. Allergic reactions are exceedingly rare, and no drug interactions have been identified for topical use.

Interactions and Precautions

No clinically relevant interactions with other substances are known when allantoin is applied topically. Because allantoin softens the stratum corneum, it may slightly enhance the skin penetration of other active ingredients in the same formulation. Individuals with known sensitivity to comfrey extracts should use products containing plant-derived allantoin with caution.
