What is Ergocalciferol?
Ergocalciferol, commonly known as vitamin D2, is the plant-derived form of vitamin D. It is produced when ergosterol -- a sterol found in fungi, yeasts, and certain plants -- is exposed to ultraviolet (UV) radiation. Along with cholecalciferol (vitamin D3), which is of animal origin or synthesized in human skin upon sun exposure, ergocalciferol is one of the two major forms of vitamin D relevant to human health.
Biological Functions
Ergocalciferol plays a crucial role in calcium and phosphate metabolism. After ingestion, it is converted in the liver to 25-hydroxyvitamin D2 and subsequently activated in the kidneys to its biologically active form, 1,25-dihydroxyvitamin D2 (calcitriol). Key biological functions include:
- Promoting calcium absorption in the intestine
- Regulating blood calcium and phosphate levels
- Supporting bone mineralization and skeletal health
- Modulating immune system function
- Contributing to muscle strength and nerve function
Dietary Sources
Ergocalciferol is primarily found in plant-based and fungal food sources. The most notable sources include:
- Mushrooms (especially UV-exposed varieties such as shiitake, button mushrooms, and chanterelles)
- Foods fortified with vitamin D2 (e.g., plant-based milks, juices, and breakfast cereals)
- Yeasts and yeast-based products
Compared to animal-derived vitamin D3 sources, the ergocalciferol content in natural foods tends to be modest. However, for individuals following a vegan or vegetarian diet, UV-exposed mushrooms and fortified products represent important dietary sources.
Recommended Intake
The recommended daily intake of vitamin D (including both ergocalciferol and cholecalciferol) varies by age, health status, and sun exposure. According to the World Health Organization (WHO) and major nutrition authorities, general guidance is as follows:
- Infants: 10 µg (400 IU) per day
- Children, adolescents, and adults: 15-20 µg (600-800 IU) per day
- Older adults (65 years and above): 20 µg (800 IU) per day
In cases of confirmed vitamin D deficiency, higher therapeutic doses may be prescribed under medical supervision.
Deficiency Symptoms
A deficiency in vitamin D2 (ergocalciferol) manifests in the same way as general vitamin D deficiency. Common symptoms and related conditions include:
- Rickets: Bone softening and skeletal deformities in children
- Osteomalacia: Bone softening in adults, associated with bone and muscle pain
- Osteoporosis: Reduced bone density and increased fracture risk
- Muscle weakness and increased risk of falls
- Impaired immune function and greater susceptibility to infections
- Fatigue and depressive mood
Risk Groups for Vitamin D Deficiency
- People with limited sun exposure (e.g., elderly individuals, office workers, shift workers)
- Individuals with darker skin tones
- Vegans and vegetarians without supplementation
- People with malabsorption disorders (e.g., Crohn's disease, celiac disease)
- Obese individuals
- Pregnant and breastfeeding women
Medical Use and Supplementation
Ergocalciferol is widely used as a pharmaceutical agent and dietary supplement to prevent or treat vitamin D deficiency. It is available in several forms:
- Tablets and capsules
- Oral drops (particularly suitable for infants)
- Injectable solutions (for severe deficiency states)
Ergocalciferol is considered safe and well-tolerated when taken at recommended doses. Some studies suggest that cholecalciferol (vitamin D3) may raise blood levels of 25-hydroxyvitamin D slightly more efficiently. Nevertheless, ergocalciferol remains an important alternative, particularly for vegans, as it is derived from plant sources.
Overdose and Toxicity
Toxicity from ergocalciferol is rare through dietary intake alone but can occur with prolonged high-dose supplementation. Symptoms of vitamin D toxicity (hypervitaminosis D) include:
- Nausea, vomiting, and loss of appetite
- Elevated blood calcium levels (hypercalcemia)
- Kidney damage and calcification
- Cardiac arrhythmias
- Muscle weakness and confusion
The tolerable upper intake level (UL) for adults, as established by the European Food Safety Authority (EFSA), is 100 µg (4,000 IU) per day. Higher doses should only be taken under medical supervision.
Interactions
Ergocalciferol may interact with certain medications and nutrients:
- Calcium: Combined intake of calcium and vitamin D enhances calcium absorption; at high doses of both, the risk of hypercalcemia may increase.
- Thiazide diuretics: May elevate blood calcium levels, increasing the risk of hypercalcemia when combined with vitamin D.
- Cholestyramine and orlistat: May impair the absorption of fat-soluble vitamins, including vitamin D.
- Antiepileptic drugs and glucocorticoids: May interfere with vitamin D metabolism and increase requirements.
