What is Genotyping?

Genotyping refers to the analysis of an organism´s genetic material to determine its specific genetic profile, known as the genotype. It involves identifying variations in DNA sequences that can provide information about hereditary traits, disease risks, or the ancestry of an individual. Genotyping is applied in clinical diagnostics, personalized medicine, pharmacogenetics, and basic research.

Methods of Genotyping

Several technical methods are used for genotyping, selected depending on the clinical or research question and the level of resolution required:

  • Polymerase Chain Reaction (PCR): One of the most widely used methods, in which specific DNA segments are amplified and subsequently analyzed.
  • DNA Sequencing (e.g., Next-Generation Sequencing, NGS): Enables complete or targeted determination of the DNA sequence, allowing the identification of mutations and variants.
  • SNP Genotyping (Single Nucleotide Polymorphism): Analysis of individual nucleotide variants in the genome that may be associated with diseases or drug responses.
  • Microarray Technology: Allows simultaneous analysis of hundreds of thousands of genetic variants on a single chip.
  • RFLP Analysis (Restriction Fragment Length Polymorphism): An older method for identifying genetic differences based on cutting patterns of specific enzymes.

Areas of Application

Clinical Diagnostics

In medicine, genotyping is used to diagnose genetic diseases such as cystic fibrosis, sickle cell anemia, or hereditary cancer syndromes. It also enables carrier detection for recessive genetic disorders and supports prenatal diagnostics.

Pharmacogenetics and Personalized Medicine

Pharmacogenetics uses genotyping to predict how a patient will respond to specific medications. Genetic variants in enzymes such as CYP2D6 or CYP2C19 influence drug metabolism and can be critical for dose adjustments. This allows therapy to be individually tailored to the patient – a central element of personalized medicine.

Infectious Diseases

In infectious diseases, genotyping of pathogens such as HIV, Hepatitis B and C, or Mycobacterium tuberculosis enables the determination of the viral or bacterial subtype (genotype). This is essential for selecting the most effective antiviral or antibiotic therapy and for assessing resistance patterns.

Oncology

In cancer medicine, genotyping of tumor cells helps identify specific mutations that serve as targets for targeted therapies. Examples include mutations in the BRCA1/2 gene in breast and ovarian cancer, or EGFR mutations in lung carcinoma.

Forensics and Ancestry Analysis

Genotyping is also used in forensics to identify individuals based on DNA traces, as well as for kinship analysis and ancestry determination.

Relevance for Patient Care

With the increasing availability and decreasing costs of modern sequencing technologies, genotyping is becoming an increasingly important tool in clinical practice. It helps to establish more precise diagnoses, tailor therapies to individual patients, and reduce the risk of adverse drug reactions. Within screening programs, it can also be used for the early detection of genetic risk factors.
