What is Malate?

Malate is the ionized form of malic acid, an organic compound found naturally in many fruits – especially apples – and various vegetables. In the human body, malate plays a central role in the citric acid cycle (also known as the Krebs cycle), which is the primary metabolic pathway for cellular energy production.

Biological Functions

Malate serves several important functions in the human body:

  • Energy metabolism: As an intermediate in the citric acid cycle, malate is directly involved in cellular ATP production. It is formed from fumarate and converted to oxaloacetate, enabling continuation of the cycle.
  • Gluconeogenesis: Malate contributes to glucose synthesis via the malate-aspartate shuttle and supports the transfer of reducing equivalents between mitochondria and the cytoplasm.
  • Detoxification: Malate participates in the urea cycle and supports the detoxification of ammonia in the liver.
  • Acid-base balance: As an organic acid, malate contributes to the regulation of the body's pH.

Dietary Sources

Malate occurs naturally in many foods, particularly in:

  • Apples and pears
  • Cherries and plums
  • Grapes
  • Tomatoes and carrots
  • Broccoli and other cruciferous vegetables

In the food industry, malic acid (E 296) is used as an acidulant and flavor enhancer. Malate salts such as sodium malate (E 350), potassium malate (E 351), and calcium malate (E 352) serve as acidity regulators in processed foods.

Malate as a Dietary Supplement

In the supplement industry, malate is often combined with minerals to enhance their bioavailability and metabolic effects:

  • Magnesium malate: A well-tolerated form of magnesium that is also thought to support energy production.
  • Citrulline malate: A combination of the amino acid citrulline and malate, commonly used in sports nutrition to improve endurance performance.

Some studies have investigated malate in relation to reducing fatigue and improving athletic performance. However, the scientific evidence for many of these applications remains limited and further research is needed.

Recommended Intake and Safety

There are no specific dietary intake recommendations for malate from natural food sources, as the body is capable of synthesizing it endogenously. As a supplement, typical dosages range from 1,000 to 2,800 mg per day, depending on the compound form and intended use.

Malate is generally considered safe (classified as GRAS – Generally Recognized As Safe in the United States). At very high doses, mild gastrointestinal discomfort may occasionally occur. Individuals with kidney disease should consult a healthcare professional before taking malate-containing supplements.

Interactions

Malate may influence the intestinal absorption of certain minerals, such as magnesium or iron, when taken in combination. No clinically significant interactions with medications have been documented to date; however, caution is advised when combining malate supplements with mineral preparations.
