Things worth knowing about "Microbiome Sequencing"

Microbiome sequencing is a modern diagnostic method that genetically analyzes all microorganisms living in or on the human body, providing key insights into health and disease.

What Is Microbiome Sequencing?

Microbiome sequencing is an advanced diagnostic technique used to analyze the genetic composition of all microorganisms found in a biological sample – most commonly from the gut, skin, or other body sites. The term microbiome refers to the complete community of microorganisms (bacteria, viruses, fungi, and archaea) that inhabit the human body. Sequencing allows researchers and clinicians to precisely characterize this community without needing to culture individual organisms in a laboratory setting.

Methods of Microbiome Sequencing

Several approaches are used to sequence the microbiome:

  • 16S rRNA Sequencing: This method targets a conserved gene (the 16S rRNA gene) present in virtually all bacteria. It offers a fast and cost-effective way to identify bacterial communities at the genus or family level.
  • Metagenomic Sequencing (Shotgun Sequencing): This comprehensive approach sequences all DNA in a sample. It provides information on bacteria, viruses, fungi, and archaea, and allows for functional profiling of the microbiome.
  • Metatranscriptomics and Metabolomics: Complementary methods that measure active genes (RNA) and metabolic byproducts of the microbiome, providing an even more detailed picture of microbial activity.

When Is Microbiome Sequencing Used?

Microbiome sequencing is applied in both research and increasingly in clinical settings. Common areas of application include:

  • Investigation of the gut microbiome in conditions such as Crohn's disease, irritable bowel syndrome, or inflammatory bowel disease
  • Analysis of the microbiome in oncology, as the gut microbiome can influence the effectiveness of cancer therapies
  • Research into obesity, type 2 diabetes, and metabolic disorders
  • Study of the vaginal microbiome in fertility issues or infections
  • Analysis of the skin microbiome in conditions such as atopic dermatitis or psoriasis
  • Personalized nutritional counseling and preventive health diagnostics

How Microbiome Sequencing Works

Typically, a stool sample (for the gut microbiome), a swab, or another biological specimen is collected and sent to a specialized laboratory. There, DNA is extracted, sequenced, and analyzed using bioinformatics tools. Results are generally presented in detailed reports that visualize the composition of microorganisms and their relative proportions.

Clinical Relevance and Scientific Insights

A balanced microbiome is closely associated with healthy immune function, digestion, mental well-being (the gut-brain axis), and metabolism. Research shows that dysbiosis – an imbalance in the microbial community – is linked to a wide range of diseases. Microbiome sequencing is an essential tool for understanding these connections and developing targeted therapies.

Limitations and Considerations

Despite its promise, microbiome sequencing currently has limitations: results can vary between laboratories and methods, and not all findings can be directly translated into clinical recommendations. Interpreting the data requires significant bioinformatic expertise. Many commercial offerings also lack standardized reference values. Patients are advised to discuss any results with a qualified healthcare professional.

Macronutrients

Macronutrients are the three main nutrients -- carbohydrates, proteins, and fats -- that provide the body with energy and support vital physiological functions.

Food craving

A food craving is a sudden, intense desire to eat specific foods, often high in sugar or fat. It can be triggered by blood sugar fluctuations, stress, or sleep deprivation.

Portion control

Portion control refers to the conscious management of food quantities consumed at meals to regulate calorie intake and support a healthy body weight.
