Visfatin is a hormone produced mainly by adipose tissue that plays a key role in metabolism and inflammatory processes in the body.

What is Visfatin?

Visfatin is an adipokine – a signaling molecule produced primarily by visceral adipose tissue (the fat surrounding the internal organs in the abdominal area) and released into the bloodstream. The name is derived from the words visceral fat and -atin. Visfatin is identical to the enzyme NAMPT (Nicotinamide Phosphoribosyltransferase), which plays a central role in cellular energy metabolism.

Biological Functions

Visfatin fulfills several important roles in the body:

  • Insulin-mimetic activity: Visfatin can bind to the insulin receptor and exert an insulin-like effect, promoting glucose uptake into cells.
  • Regulation of inflammation: Visfatin acts as a cytokine and can stimulate pro-inflammatory processes throughout the body.
  • NAD+ biosynthesis: As the NAMPT enzyme, visfatin catalyzes a key step in the synthesis of nicotinamide adenine dinucleotide (NAD+), an essential coenzyme for cellular metabolism.
  • Immunomodulation: Visfatin influences immune cells such as macrophages and neutrophils, altering their activity and function.

Visfatin and Disease

Obesity and Metabolic Syndrome

In overweight and obese individuals, blood visfatin levels are frequently elevated. This has been linked to the metabolic syndrome – a cluster of conditions including abdominal obesity, high blood pressure, elevated blood lipids, and insulin resistance.

Type 2 Diabetes

Elevated visfatin levels have been observed in patients with type 2 diabetes mellitus. However, the precise role of visfatin in the development and progression of this disease remains an active area of research.

Cardiovascular Disease

Studies suggest that high visfatin levels may be associated with an increased risk of atherosclerosis (hardening of the arteries) and cardiovascular disease.

Chronic Inflammatory Diseases

Because visfatin has pro-inflammatory properties, it has also been investigated in conditions such as rheumatoid arthritis, inflammatory bowel disease, and other autoimmune disorders.

Oncology

Emerging research suggests that visfatin may play a role in tumor growth, as it can promote cell proliferation and survival. Elevated visfatin levels have been observed in various types of cancer.

Measurement and Diagnostics

Visfatin can be measured in blood serum using an ELISA (Enzyme-Linked Immunosorbent Assay). Elevated visfatin levels are considered a potential biomarker for metabolic disorders and chronic inflammation. However, routine clinical measurement of visfatin is not yet standardized and remains primarily within the scope of scientific research.

Factors Influencing Visfatin Levels

Several factors can affect visfatin concentrations in the blood:

  • Body weight: Increasing visceral fat leads to higher visfatin levels.
  • Diet: A calorie-rich, unbalanced diet can increase visfatin production.
  • Physical activity: Regular exercise may help reduce visfatin levels.
  • Age and sex: Visfatin concentrations can vary depending on age and biological sex.
  • Medications: Certain drugs, such as some antidiabetic agents, may influence visfatin levels.
