What is Neotame?
Neotame (EU food additive number: E961) is an artificial sweetener belonging to the group of high-intensity sweeteners. It is a derivative of the well-known sweetener aspartame, but has been chemically modified to provide a significantly greater sweetening power. Neotame is approximately 7,000 to 13,000 times sweeter than table sugar (sucrose) and is therefore needed only in very small amounts. In the European Union, neotame is approved as a food additive and may be used in a wide range of foods and beverages.
Chemical Properties and Mechanism of Action
Chemically, neotame is an N-neohexyl derivative of aspartame. Despite its structural similarity to aspartame, neotame is not metabolized into phenylalanine in the body. This makes it potentially safer -- unlike aspartame -- for individuals with phenylketonuria (PKU), although individual medical advice is still recommended.
Its sweetening effect results from the highly specific binding to sweet taste receptors (T1R2/T1R3) on the tongue. Because it is used in extremely small quantities and undergoes minimal metabolism, neotame provides virtually no calories.
Applications and Occurrence
Neotame is used as a sweetening agent and flavor enhancer in various food categories. Typical applications include:
- Calorie-reduced and sugar-free soft drinks
- Chewing gums and confectionery
- Dairy products and desserts
- Baked goods and breakfast cereals
- Diet products and dietary supplements
- Tabletop sweeteners
Due to its enormous sweetening power, only microgram-to-milligram quantities are required per serving.
Safety and Health Assessment
Neotame has been evaluated by several international food safety authorities and has been classified as safe:
- EFSA (European Food Safety Authority): Approved neotame as E961 and established an ADI (Acceptable Daily Intake) of 2 mg per kg of body weight per day.
- FDA (U.S. Food and Drug Administration): Classified neotame as GRAS (Generally Recognized As Safe).
- JECFA (Joint FAO/WHO Expert Committee on Food Additives): Also evaluated neotame as safe for use in food.
Since neotame is used in extremely low concentrations, exceeding the ADI under normal dietary conditions is considered very unlikely.
Possible Side Effects and Criticism
As with other synthetic sweeteners, there is ongoing public debate about potential side effects. However, no convincing scientific evidence of harmful effects at approved usage levels currently exists. Some studies have generally investigated whether high-intensity sweeteners might influence the gut microbiome or insulin metabolism. The data specifically on neotame remains limited, and further research is desirable.
Individuals with hypersensitivities to aspartame-related compounds should exercise caution when consuming products containing neotame and consult a healthcare professional if in doubt.
Labeling
In the European Union, foods containing neotame must declare this additive in the ingredient list either by its name Neotame or by its designation E961. Unlike aspartame, neotame does not require a special warning label for individuals with PKU under current EU regulations.
