The nitro group is a chemical functional group with the general formula -NO2, consisting of one nitrogen atom (N) bonded to two oxygen atoms (O). It is covalently attached to an organic molecule and fundamentally alters its chemical and biological properties. Compounds bearing one or more nitro groups are referred to as nitro compounds.
Chemical Properties
The nitro group is strongly electron-withdrawing. It pulls electron density away from the molecule, affecting its reactivity and polarity. Key chemical features include:
- Resonance: The nitro group exists in resonance structures in which the negative charge is delocalized across both oxygen atoms.

They are metabolized in the body to nitric oxide (NO), which relaxes the smooth muscle of blood vessel walls, resulting in vasodilation. This is especially important in the treatment of angina pectoris and heart failure.
Antibiotics and Anti-infective Agents
Several antibiotics and antiparasitic drugs contain a nitro group:
- Metronidazole: A nitroimidazole antibiotic active against anaerobic bacteria and protozoa. The nitro group is reduced intracellularly, generating reactive intermediates that damage the DNA of the pathogens.
- Nitrofurantoin: Used to treat urinary tract infections, its mechanism also depends on the reduction of the nitro group to toxic reactive species that kill bacteria.
- Chloramphenicol: A broad-spectrum antibiotic that likewise carries a nitro group in its molecular structure.
Other Drugs Containing a Nitro Group
- Nilutamide: An antiandrogen used in the treatment of prostate cancer.
- Nifedipine and other dihydropyridine calcium channel blockers also contain a nitro group and are used for hypertension and cardiac conditions.
Toxicological Significance
Nitro compounds can be toxic. Certain industrial nitroaromatics such as nitrobenzene or nitrotoluene are poisonous and can cause methemoglobinemia upon exposure -- a condition in which hemoglobin is unable to carry oxygen effectively. Symptoms include bluish discoloration of the skin (cyanosis), dizziness, and shortness of breath.
Significance in Explosives Chemistry
Nitro groups are a defining structural feature of many explosive compounds. Substances such as trinitrotoluene (TNT), nitroglycerin, and RDX contain multiple nitro groups. The rapid decomposition of these compounds with the release of large volumes of gas is the basis of their explosive power.
Significance in Environmental Chemistry
Nitroaromatic compounds can occur as environmental contaminants in soil and water. They are produced, among other sources, by industrial processes and the combustion of fossil fuels. Due to their stability and potential toxicity, they are considered problematic environmental pollutants.
