What is a Neuropeptide?
A neuropeptide is a short chain of amino acids (a peptide) produced and released within the nervous system. Neuropeptides function as chemical messengers, transmitting signals between nerve cells (neurons) and between the nervous system and other organs. Unlike classical neurotransmitters such as serotonin or dopamine, neuropeptides are larger in molecular size and typically exert more prolonged and wide-ranging effects.
More than 100 distinct neuropeptides have been identified in the human body. They play a central role in regulating a wide variety of physiological and psychological processes, including pain perception, hunger, sleep, mood, stress responses, and memory.
Structure and Production
Neuropeptides are composed of short chains of amino acids, the building blocks of proteins. They are synthesized in nerve cells from larger precursor proteins known as prepropeptides, stored in vesicles, and released upon stimulation. Once released, they bind to specific receptors on target cells, triggering a biological response.
Key Neuropeptides and Their Functions

- Chronic pain conditions: Altered substance P signaling can contribute to pain hypersensitivity.
- Depression and anxiety disorders: Imbalances in neuropeptides such as corticotropin-releasing hormone (CRH) have been linked to psychiatric conditions.
- Obesity and eating disorders: The neuropeptide Y system is closely involved in the regulation of hunger and body weight.
- Migraine: Calcitonin gene-related peptide (CGRP) plays a pivotal role in migraine pain; modern migraine medications specifically target this neuropeptide.
Medical and Therapeutic Relevance
Research into neuropeptides has significant implications for drug development. CGRP-based therapies, including CGRP antagonists, represent a major advance in migraine treatment. Neuropeptides are also being investigated as therapeutic targets in pain management, oncology, and psychiatry.
