What is Butyl?
Butyl refers to an organic chemical group (alkyl group) with the molecular formula C₄H₉, derived from butane. Depending on the structure, four isomers are distinguished: n-butyl (normal butyl), iso-butyl, sec-butyl (secondary butyl), and tert-butyl (tertiary butyl). Butyl groups are components of numerous chemical compounds and are widely used in industry, pharmacology, and everyday products.
Chemical Properties
Butyl groups consist of four carbon atoms and nine hydrogen atoms. They belong to the class of alkyl groups, which are saturated hydrocarbon residues. Their properties vary depending on the isomer:
- n-Butyl: linear chain, highly reactive
- iso-Butyl: branched chain, slightly less reactive
- sec-Butyl: secondary carbon atom as the point of attachment
- tert-Butyl: tertiary carbon atom, sterically demanding, particularly stable

- Butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA): synthetic antioxidants used as food additives (E320, E321)
- Parabens (e.g., butylparaben): preservatives used in cosmetics and pharmaceutical products
Pharmaceutical Relevance
In the field of pharmacy, butyl compounds play an important role:
- Butylscopolamine (scopolamine-N-butyl bromide): an antispasmodic agent used to relieve abdominal cramps and gastrointestinal complaints
- tert-Butyl groups are used in drug synthesis to improve the stability and bioavailability of molecules
- Butyl rubber is used for closures of injection vials and medical packaging
Health Aspects and Safety
The health assessment of butyl compounds varies greatly depending on the specific compound:
- Butylparaben: is subject to ongoing debate as it is considered weakly estrogenic. The European Food Safety Authority (EFSA) and the Scientific Committee on Consumer Safety (SCCS) have established maximum permitted levels.
- BHA and BHT: are classified as safe in approved quantities, but are under observation at high doses.
- Butyl acetate: can cause irritation of the respiratory tract, eyes, and skin upon heavy exposure.
- Butylscopolamine: is an over-the-counter medication and is considered safe when used as directed.
Regulation and Labeling
Butyl compounds are subject to various regulatory requirements depending on their area of use. In the European Union, butyl-based food additives (BHA as E320, BHT as E321) are subject to mandatory labeling. Cosmetic products containing butylparaben must list this substance in the ingredient list (INCI). Medicinal products containing butyl compounds are subject to authorization by the relevant regulatory authorities (e.g., EMA, BfArM).
