A functional group is a specific atom or group of atoms within an organic molecule that is responsible for the characteristic chemical and physical properties of that molecule. Functional groups determine how a molecule reacts with other substances, how it dissolves in water, and what biological effects it produces. In medicine and biochemistry, functional groups are of central importance because they govern the interactions of drugs, enzymes, hormones, and other biomolecules with their target structures in the body.
Importance in Medicine and Biochemistry
In medical and pharmaceutical contexts, functional groups play a decisive role. The chemical structure of a drug -- particularly its functional groups -- determines whether it is absorbed, metabolized, or excreted by the body. Binding to receptors and enzymes is also significantly influenced by functional groups.
- Pharmacology: Functional groups determine the lipophilicity or hydrophilicity of a drug, affecting its ability to cross cell membranes and its bioavailability.
- Enzyme reactions: Enzymes recognize their substrates through specific functional groups and catalyze biochemical reactions in metabolism.
- Receptor binding: The interaction between a drug and its receptor is based on complementary functional groups that form hydrogen bonds, ionic interactions, or hydrophobic contacts.
Key Functional Groups in Biochemistry
Numerous functional groups occur in biological molecules, each conferring specific properties:
- Hydroxyl group (-OH): Found in alcohols, sugars, and many hormones. It makes molecules water-soluble and allows hydrogen bonding.
- Carboxyl group (-COOH): Characteristic of amino acids and fatty acids. It gives the molecule acidic properties.
- Amino group (-NH₂): Found in amino acids, neurotransmitters, and nucleotides. It acts as a base and is essential for peptide bond formation.
- Phosphate group (-PO₄): Important in energy transfer (ATP) and as a structural component of DNA and RNA.
- Carbonyl group (C=O): Found in aldehydes and ketones, for example in sugar molecules such as glucose.
- Ester group (-COO-): Typical of fats (triglycerides) and many drugs such as aspirin.
- Sulfhydryl group (-SH): Important for protein folding through disulfide bridges, for example in cysteine.
Functional Groups and Drug Development
In medicinal chemistry and drug design, functional groups are deliberately manipulated. By replacing or modifying individual functional groups, the potency, selectivity, stability, and tolerability of a drug candidate can be optimized. This principle is known as structure-activity relationship (SAR). For example, introducing a hydroxyl group can increase the water solubility of a drug, while adding a fluorine atom can improve metabolic stability.
Relevance to Metabolism
In human metabolism, functional groups are chemically altered by enzymes to activate, deactivate, or prepare substances for excretion. The so-called Phase I metabolism in the liver often involves the introduction or unmasking of functional groups (e.g., hydroxylation by cytochrome P450 enzymes), while in Phase II metabolism, functional groups are conjugated with endogenous molecules to facilitate elimination from the body.
