Nicotinamide Riboside (NR) is a naturally occurring form of Vitamin B3 and a key precursor to NAD+, a coenzyme essential for cellular energy production and overall metabolic health.
What is Nicotinamide Riboside (NR)?
Nicotinamide Riboside (NR) is a unique form of Vitamin B3 (niacin) belonging to the family of nicotinamide nucleotides. It is found in trace amounts in certain foods such as cows milk, yeast, and some vegetables. In recent years, NR has gained significant scientific and public attention as a dietary supplement, primarily because it serves as a direct precursor to Nicotinamide Adenine Dinucleotide (NAD+) -- a coenzyme that plays a critical role in virtually every cell of the human body.
Mechanism of Action
After ingestion, NR is rapidly converted in the body first into Nicotinamide Mononucleotide (NMN) and then into NAD+. NAD+ is involved in hundreds of biochemical reactions, including:
- Cellular respiration and mitochondrial energy production
- Activation of sirtuins (proteins that regulate cell repair and promote longevity)
- Activation of PARP enzymes involved in DNA repair
- Regulation of inflammation and metabolic processes
NAD+ levels naturally decline with age, and this decline is associated with reduced physical performance, metabolic disease, and increased susceptibility to age-related conditions. NR supplementation aims to restore and elevate NAD+ levels to support these vital biological functions.
Medical Uses and Health Benefits
Energy Metabolism and Physical Performance
Because NAD+ is essential for mitochondrial energy production, NR is used to combat fatigue and support both physical and cognitive performance. Early clinical trials have demonstrated that NR supplementation can significantly increase NAD+ levels in the blood of healthy adults.
Aging and Cellular Renewal
NR is actively investigated in the field of longevity research. By activating sirtuins and promoting mitochondrial biogenesis, NR may contribute to healthy aging at the cellular level. Animal studies have shown promising results, though robust human clinical data is still emerging.
Neuroprotection and Cognitive Function
There is growing evidence that elevated NAD+ levels may exert neuroprotective effects. NR is being studied in the context of preventing or slowing neurodegenerative conditions such as Alzheimer disease and Parkinson disease, although large-scale human trials are still needed.
Cardiovascular Health
Preclinical studies suggest that NR may have beneficial effects on blood pressure, vascular function, and heart health. One clinical study reported a reduction in arterial blood pressure among older adults who supplemented with NR.
Metabolic Health and Diabetes Prevention
NR may improve insulin sensitivity and support healthy lipid metabolism. Animal studies have shown protective effects against obesity and type 2 diabetes, and initial human studies offer encouraging preliminary findings.
Dietary Sources
NR is found in small amounts in the following foods:
- Cows milk and dairy products
- Yeast (e.g., brewers yeast)
- Certain vegetables such as broccoli and edamame
- Meat and fish (in very small quantities)
However, the natural dietary intake of NR is very low, and therapeutically relevant amounts are generally only achievable through supplementation.
Dosage and Administration
Clinical studies have commonly used doses ranging from 250 mg to 1000 mg per day. Standard supplements typically contain 250 mg or 300 mg of NR per capsule. It is generally recommended to take NR in the morning, as it may support alertness and energy levels throughout the day. No official recommended daily intake has been established by regulatory bodies such as the EFSA or WHO to date.
Safety and Side Effects
NR is considered well-tolerated and safe at the doses studied in clinical trials to date. No serious adverse events have been reported. Mild side effects may occasionally occur, including:
- Nausea or mild gastrointestinal discomfort
- Headaches
- Fatigue (rare)
- Skin flushing (less common than with other niacin forms)
Pregnant or breastfeeding individuals and those with pre-existing medical conditions should consult a healthcare professional before starting NR supplementation. Long-term human safety data is still limited.
Interactions
NR may potentially interact with certain medications, including:
- Chemotherapy agents: Since NR can enhance DNA repair mechanisms, its use during cancer treatment is a topic of ongoing discussion and caution.
- Anticoagulants: Potential interactions with blood-thinning medications have not been fully clarified.
Concurrent use of other NAD+ precursors such as NMN or high doses of other Vitamin B3 forms should be discussed with a physician.
