What Are Fatty Acid Methyl Esters?
Fatty acid methyl esters (FAME) are organic chemical compounds produced by reacting fatty acids with methanol. During this reaction, the carboxyl group of the fatty acid combines with the hydroxyl group of methanol, releasing water and forming an ester bond. This process is called esterification or, when triglycerides serve as the starting material, transesterification.
Fatty acid methyl esters do not occur in significant amounts in nature but are deliberately synthesized in laboratory and industrial settings. They play an important role in food analysis, pharmacology, cosmetics, and as biofuel.
Chemical Properties
FAME are derived from naturally occurring fatty acids found in vegetable oils, animal fats, and fish oils. The properties of the methyl esters vary considerably depending on the parent fatty acid:
- Saturated fatty acid methyl esters (e.g., methyl palmitate, methyl stearate): solid to waxy at room temperature, high melting point.
- Monounsaturated fatty acid methyl esters (e.g., methyl oleate): liquid, low melting point.
- Polyunsaturated fatty acid methyl esters (e.g., methyl linoleate, methyl-EPA, methyl-DHA): liquid, sensitive to oxidation.
Compared to free fatty acids, methyl esters are less reactive, more volatile, and easier to separate chromatographically, making them particularly suitable for analytical purposes.
Production
FAME are typically produced by two methods:
- Direct esterification: Free fatty acids are reacted with methanol in the presence of an acid catalyst (e.g., sulfuric acid).
- Transesterification: Triglycerides (e.g., from vegetable oils) react with methanol under alkaline or acid catalysis to yield fatty acid methyl esters and glycerol. This process is the basis of biodiesel production.
Fields of Application
Food Analysis and Nutritional Research
In analytical chemistry, fatty acids from food, blood plasma, or tissue samples are first converted into their methyl esters to enable identification and quantification by gas chromatography (GC). This method is the gold standard for determining the fatty acid profile of food products and biological samples, allowing precise measurement of omega-3 and omega-6 fatty acid content in supplements and food items.
Pharmacology and Medicine
In pharmaceutical science, fatty acid methyl esters are used as starting materials or intermediates in the production of omega-3 fatty acid preparations (e.g., EPA and DHA). Highly purified omega-3 ethyl esters and methyl esters are used in the treatment of hypertriglyceridemia (elevated blood triglyceride levels). It should be noted that the bioavailability of fatty acids in methyl ester form may differ compared to triglycerides or free fatty acids.
Cosmetics and Pharmaceutical Excipients
Fatty acid methyl esters are used as emollients (skin-conditioning agents), solvents, and carrier substances in creams, lotions, and pharmaceutical formulations. They improve the texture of skin care products and support the penetration of active ingredients into the skin.
Biofuel (Biodiesel)
The most well-known industrial application of FAME is their use as biodiesel. The transesterification of vegetable oils (e.g., rapeseed oil, soybean oil, palm oil) with methanol produces a fuel suitable for diesel engines. FAME-based biodiesel is biodegradable and classified as a renewable energy source.
Health Relevance and Safety
Fatty acid methyl esters are generally used in analytical and industrial quantities and are not directly used as food additives. Some methyl esters, particularly those derived from short-chain fatty acids, can be irritating to skin and mucous membranes and should not be inhaled or ingested in larger quantities. In pharmaceutical and cosmetic applications, purified fatty acid methyl esters are considered safe when used as intended.
When ingested orally, fatty acid methyl esters are hydrolyzed in the gastrointestinal tract, releasing free fatty acids and methanol. The small amounts of methanol produced during normal dosage are toxicologically harmless.
