What is cAMP?
cAMP (cyclic adenosine monophosphate, also written as cyclic AMP) is a small but critically important molecule found in virtually all living cells. It acts as a second messenger -- a molecule that relays signals from hormones or neurotransmitters on the cell surface to the interior of the cell. cAMP is synthesized from adenosine triphosphate (ATP) by the enzyme adenylyl cyclase and plays a central role in intracellular signal transduction.
Biochemical Basis
The production of cAMP is triggered when a hormone or signaling molecule (for example, adrenaline) binds to a G protein-coupled receptor (GPCR) on the cell surface. This activates adenylyl cyclase, which converts ATP into cAMP. cAMP then activates key intracellular enzymes, most notably protein kinase A (PKA), which phosphorylates target proteins and triggers a wide range of cellular responses.
cAMP is broken down by enzymes called phosphodiesterases (PDEs), which convert it into the inactive 5´-AMP. This degradation step ensures that signals are properly terminated and cellular responses remain tightly controlled.
Biological Functions
- Hormone signaling: cAMP mediates the effects of hormones such as adrenaline, glucagon, thyroid-stimulating hormone (TSH), and adrenocorticotropic hormone (ACTH).
- Metabolic regulation: It promotes glycogen breakdown (glycogenolysis) and fat breakdown (lipolysis), increasing energy availability during stress or exercise.
- Cardiac function: cAMP increases heart rate and the force of heart contractions (positive chronotropy and inotropy).
- Immune modulation: It regulates inflammatory responses and the activity of immune cells such as T-cells and macrophages.
- Nervous system: cAMP is involved in neurotransmitter release, neuronal plasticity, and long-term memory formation.
- Gene expression: Via the transcription factor CREB (cAMP response element-binding protein), cAMP influences the expression of specific genes.
Clinical Relevance
Dysregulation of the cAMP signaling pathway is linked to a range of diseases. Excessive activation, for example due to activating mutations in the Gs protein, underlies conditions such as McCune-Albright syndrome. The toxin produced by Vibrio cholerae (cholera toxin) dramatically elevates cAMP levels in intestinal cells, causing the profuse watery diarrhea characteristic of cholera.
Conversely, the cAMP pathway is a major therapeutic target:
- Phosphodiesterase inhibitors (e.g., sildenafil, theophylline, milrinone) raise cAMP levels by blocking its breakdown and are used in conditions such as erectile dysfunction, asthma, and heart failure.
- Beta-2 agonists (e.g., salbutamol) activate cAMP signaling in airway smooth muscle, causing bronchodilation in asthma treatment.
- Adrenaline, used in emergencies, harnesses the cAMP pathway to boost cardiac output and relax bronchial muscle.
cAMP in Research
cAMP remains one of the most intensively studied molecules in pharmacology and cell biology. Novel drugs targeting the cAMP pathway are being developed for cardiovascular disease, diabetes, inflammatory conditions, and neurological disorders. Fluorescence-based biosensors now allow real-time measurement of cAMP dynamics in living cells, opening new avenues for drug discovery.
