What Is a Capsid?
The capsid is the outer protein shell of a virus. It surrounds and protects the viral genetic material – either DNA or RNA – from degradation by enzymes or chemical agents in the environment. The capsid is an essential component of every virus particle, also referred to as a virion.
Structure of the Capsid
The capsid is composed of identical or similar protein subunits called capsomeres. These subunits self-assemble into a stable three-dimensional structure. Depending on the virus type, the capsid can take different shapes:
- Icosahedral symmetry: The protein shell is approximately spherical and consists of 20 equilateral triangular faces. Many viruses pathogenic to humans, such as adenoviruses, adopt this shape.
- Helical symmetry: The capsomeres arrange themselves in a spiral around the nucleic acid. A well-known example is the tobacco mosaic virus.
- Complex symmetry: Some viruses, such as bacteriophages, have a more complex structure that does not fit neatly into either category.
Functions of the Capsid
The capsid serves several important biological functions:
- Protection of nucleic acid: It shields the viral genome from degradation by cellular enzymes known as nucleases.
- Host cell recognition and infection: Specific surface proteins on the capsid allow the virus to bind to receptors on the surface of host cells, which is the critical first step in initiating an infection.
- Release of genetic material: Once inside the host cell, the capsid disassembles in a controlled manner, releasing the viral nucleic acid for replication.
Capsid and Viral Envelope
Not all viruses possess an additional lipid membrane surrounding the capsid. This leads to an important distinction:
- Non-enveloped viruses (naked viruses): These have only the capsid as their outer layer. They tend to be more resistant to environmental conditions such as desiccation and certain disinfectants.
- Enveloped viruses: In these viruses, the capsid is surrounded by a viral envelope derived from the host cell membrane. These viruses are generally more sensitive to alcohol-based disinfectants and other lipid-dissolving agents.
Relevance in Medicine and Research
The capsid plays an important role in modern medicine. Because it carries virus-specific structures, it is a key target for several medical applications:
- Vaccine development: Capsid proteins can be used as antigens to stimulate an immune response without the need for infectious virus particles, forming the basis of many subunit vaccines.
- Antiviral therapies: Certain drugs known as capsid inhibitors block the assembly or disassembly of the capsid, thereby inhibiting viral replication. Capsid inhibitors have been developed for the treatment of HIV, for example.
- Gene therapy: Modified viral capsids – particularly those of adeno-associated viruses (AAV) – are used in gene therapy to deliver therapeutic genes precisely into human cells.
