What is E464?
E464, also known as hydroxypropyl methylcellulose (HPMC) or hypromellose, is a semi-synthetic food additive derived from plant-based cellulose. It is produced by chemically modifying cellulose, a natural structural component found in plant cell walls. E464 is approved as a food additive in the European Union and is considered safe for human consumption.
Origin and Production
E464 is obtained from cellulose of plant origin, typically sourced from wood pulp or cotton fibres. Through chemical treatment with propylene oxide and methyl chloride, hydroxypropyl and methyl groups are introduced into the cellulose structure, giving the compound its unique functional properties. The final product is a white or off-white powder that dissolves in cold water to form a clear, viscous gel.
Uses in Food
E464 is used in a wide variety of food products, fulfilling several important technological functions:
- Thickener: Increases viscosity in sauces, dressings, and soups
- Gelling agent: Forms gels in low-fat or vegan products
- Stabiliser: Stabilises emulsions and foams
- Humectant: Extends shelf life and freshness in baked goods
- Film-forming agent: Used to coat tablets and capsules in the pharmaceutical industry
Common food products containing E464 include baked goods, desserts, ice cream, sauces, salad dressings, meat substitutes, and gluten-free products, where it replaces the binding function normally provided by gluten.
Special Property: Thermoreversible Gelation
One of the most distinctive properties of E464 is its thermoreversible gelling behaviour: unlike most gelling agents, HPMC forms a gel upon heating and returns to a liquid state upon cooling. This unusual characteristic makes it particularly valuable in food processing, for example in the production of low-fat products where it mimics the functional role of fat during cooking.
Uses in the Pharmaceutical Industry
Beyond food, E464 plays an important role in the pharmaceutical industry. It is widely used as a tablet coating material (film coating), as a binder in tablet formulations, and as a viscosity modifier in eye drops. In ophthalmology, hypromellose is a common ingredient in artificial tear preparations used to treat dry eye syndrome.
Safety and Tolerability
E464 is considered safe by the European Food Safety Authority (EFSA) and the World Health Organization (WHO). It is not significantly absorbed by the body and is largely excreted unchanged. It is:
- non-allergenic
- suitable for vegans and vegetarians
- gluten-free
- calorie-free (non-digestible)
Because E464 is not digested, very large quantities may have a laxative effect. However, no adverse effects are known at typical dietary intake levels. No numerical ADI (Acceptable Daily Intake) value has been established for E464, as the substance is classified as safe without restriction.
Labelling and Regulation
In the European Union, E464 is regulated under Regulation (EC) No 1333/2008 on food additives. It is permitted in many food categories, often under the designation quantum satis (as much as is technologically necessary). On ingredient lists, it may appear as E464 or by its full name hydroxypropyl methylcellulose.
