What Are Sirtuins?
Sirtuins are an evolutionarily highly conserved family of NAD+-dependent deacylases and ADP-ribosyltransferases — enzymes that remove or transfer specific chemical groups on proteins. In humans, seven sirtuins have been identified, designated SIRT1 through SIRT7. They are found in different cellular compartments: the nucleus, the cytoplasm, and the mitochondria.
The name derives from the term Silent Information Regulator, as sirtuins were originally discovered as regulators of gene expression. Today, their roles are understood to be far more extensive and wide-ranging.
Mechanism of Action
Sirtuins require the molecule NAD+ (nicotinamide adenine dinucleotide) as a cofactor to perform their enzymatic activity. They catalyze the removal of acetyl groups from target proteins — a process called deacetylation. By modifying these proteins, sirtuins influence a broad range of cellular processes.
- SIRT1: Regulates inflammatory pathways, insulin sensitivity, and the activity of the tumor suppressor p53.
- SIRT2: Involved in cell cycle control and genomic stability.
- SIRT3, SIRT4, SIRT5: Located in the mitochondria; regulate energy metabolism and protect against oxidative stress.
- SIRT6: Plays a key role in DNA repair and telomere maintenance.
- SIRT7: Involved in transcriptional regulation and ribosomal RNA synthesis.
Biological Significance
Aging and Longevity
Sirtuins are at the forefront of aging research. Studies in model organisms such as yeast, nematodes, and fruit flies demonstrate that increased sirtuin activity can extend lifespan. In humans, the precise role in the aging process remains an active area of research. A critical factor is the cellular NAD+ level, which declines with age and thereby reduces sirtuin activity.
Metabolic Regulation
Sirtuins, particularly SIRT1 and SIRT3, regulate energy metabolism and enhance the body's response to caloric restriction and fasting. They promote fat oxidation, improve mitochondrial function, and can increase insulin sensitivity.
DNA Repair and Genomic Stability
SIRT1 and SIRT6 are critically involved in recognizing and repairing DNA damage. Reduced sirtuin activity can lead to an accumulation of mutations, potentially contributing to increased cancer risk.
Anti-Inflammatory Effects
By modulating the transcription factor NF-kB — a central driver of chronic inflammation — sirtuins exert anti-inflammatory effects. This makes them potential therapeutic targets for chronic inflammatory diseases.
Activating Sirtuins
Since sirtuin activity depends on NAD+ levels, various strategies are being investigated to boost this activity:
- Caloric restriction and intermittent fasting: Raise NAD+ levels and thereby enhance sirtuin activity.
- Exercise and physical activity: Promote metabolism and increase NAD+ production.
- NAD+ precursors: Compounds such as nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN) are available as dietary supplements to boost NAD+ levels.
- Resveratrol: A polyphenol found in red wine that is discussed as a SIRT1 activator, though scientific evidence in humans remains limited.
Clinical Relevance and Therapeutic Perspectives
Due to their diverse regulatory roles, sirtuins are considered promising therapeutic targets for the development of new treatments for type 2 diabetes, neurodegenerative diseases such as Alzheimer disease and Parkinson disease, cardiovascular diseases, and various cancers. Several sirtuin modulators are currently in preclinical and early clinical trials. However, broad therapeutic application has yet to be established.
