What Is E216 (Propylparaben)?
E216 is the E-number designation for Propylparaben (chemical name: propyl 4-hydroxybenzoate), a synthetic preservative belonging to the paraben family. Parabens are esters of p-hydroxybenzoic acid and have been used for decades as antimicrobial agents in food products, cosmetics, and pharmaceutical preparations. E216 prevents the growth of mold, yeast, and certain bacteria, thereby extending the shelf life of products.
Legal Status in the European Union
In the European Union, E216 (Propylparaben) and E217 (Sodium Propylparaben) have been no longer permitted as food additives since 2006. The European Food Safety Authority (EFSA) did not renew their authorization due to concerns about the hormonal activity of these compounds. In other regions, particularly the United States, propylparaben continues to be permitted in certain food products under regulated conditions.
Occurrence and Use
Historically, E216 was used in a variety of food products, including:
- Baked goods and pastries
- Jams, jellies, and marmalades
- Confectionery and chocolate products
- Deli salads and seafood products
- Flavorings and flavoring preparations
Propylparaben continues to be widely used today in cosmetics (e.g., creams, lotions, shampoos) and pharmaceutical products (e.g., as a preservative in ointments and syrups), as these applications are governed by different regulatory frameworks.
Mechanism of Action
Propylparaben exerts its antimicrobial effect by damaging the cell membranes of microorganisms and inhibiting their enzyme systems. This effectively suppresses the growth of molds, yeasts, and gram-positive bacteria. Its antimicrobial activity is pH-dependent and is most effective in the acidic to neutral pH range.
Health Assessment and Safety Concerns
The primary health concern associated with E216 relates to its estrogenic activity. Propylparaben can bind to estrogen receptors and trigger hormone-like effects in the human body, which is why it is classified as an endocrine disruptor. The following risks are discussed in the scientific literature:
- Hormonal disruption: Potential interference with the hormonal system, particularly in children and pregnant women.
- Reproductive toxicity: Animal studies have shown evidence of negative effects on sperm production and male fertility.
- Tissue accumulation: Parabens have been detected in human breast tissue, although a direct causal link to disease has not yet been conclusively established.
- Allergies and contact dermatitis: Topical application of propylparaben can cause skin irritation and allergic reactions.
Due to these concerns, EFSA reduced the acceptable daily intake (ADI) for propylparaben to 0–0.1 mg per kg of body weight and withdrew its authorization as a food additive.

Consumers can identify propylparaben on product labels under the following designations:
