What is GHRP-6?
GHRP-6 (Growth Hormone Releasing Peptide-6) is a synthetic hexapeptide belonging to the class of growth hormone secretagogues (GHS). It consists of six amino acids and was originally developed in scientific research to stimulate the natural release of growth hormone (GH) from the pituitary gland. GHRP-6 mimics the action of the endogenous peptide hormone ghrelin and binds to the same receptor, known as the GHS receptor type 1a (GHSR-1a).
Mechanism of Action
GHRP-6 exerts its effects primarily through two mechanisms:
- Binding to the GHSR-1a receptor: Activation of this receptor in the pituitary gland directly stimulates the release of growth hormone (somatotropin).
- Inhibition of somatostatin: GHRP-6 reduces the activity of somatostatin, an inhibitory hormone that normally suppresses GH secretion. This further amplifies GH release.
Compared to other GHRP compounds such as GHRP-2, GHRP-6 also significantly stimulates appetite, as ghrelin receptors in the hypothalamus are involved in regulating food intake. This effect is particularly pronounced with GHRP-6.
Medical Application and Research Context
GHRP-6 is currently used primarily in scientific and preclinical research. Potential clinical applications under investigation include:
- Growth hormone deficiency: Explored as a diagnostic tool or therapeutic approach in GH deficiency states.
- Cachexia and muscle wasting: Due to its appetite-stimulating and anabolic properties, GHRP-6 is being researched as a potential agent in disease-related weight loss.
- Cardioprotection: Preclinical studies suggest possible cytoprotective effects on cardiac tissue, particularly following ischemia-reperfusion injury.
- Wound healing: Some research investigates the influence of GHRP-6 on tissue repair and inflammatory processes.
Important: GHRP-6 is not approved as a pharmaceutical drug in most countries and may not be marketed for human use outside of clinical trials. It is frequently misused in the context of competitive sports and bodybuilding, which is both medically and legally problematic.
Dosage and Usage Notes (Research Context)
Various dosages have been examined in scientific studies. Typical dose ranges in animal studies and early clinical trials are between 1 and 2 micrograms per kilogram of body weight, administered subcutaneously or intravenously. There is no officially recommended human dose, as GHRP-6 is not approved as a medication. Any application outside of controlled studies must be considered experimental and potentially hazardous to health.
Side Effects and Risks
The following adverse effects are known or suspected from clinical and preclinical investigations:
- Strong increase in appetite: This is one of the most common and pronounced side effects of GHRP-6.
- Water retention (edema): Elevated GH levels can lead to fluid retention in tissues.
- Elevated cortisol levels: GHRP-6 can stimulate the secretion of cortisol and prolactin.
- Tingling and numbness: Particularly in the extremities.
- Insulin resistance: Chronically elevated GH levels can impair glucose metabolism.
- Potential tumor growth promotion: Since growth hormone promotes cell proliferation, there is a theoretical increased risk in individuals with pre-existing tumors.
Distinction from Similar Substances
GHRP-6 belongs to the family of GH secretagogues and should be distinguished from the following substances:
- GHRP-2: Similar mechanism of action, but less appetite stimulation and more potent GH secretion.
- Ipamorelin: A more selective GH secretagogue with fewer effects on cortisol and prolactin.
- Sermorelin / CJC-1295: GHRH analogues that act through a different receptor pathway.
- Ghrelin: The endogenous peptide hormone whose action GHRP-6 mimics.
Legal Classification
In most countries, GHRP-6 is classified as an unapproved substance without a valid pharmaceutical marketing authorization. The World Anti-Doping Agency (WADA) has placed GHRP-6 on its list of prohibited substances (category: peptide hormones, growth factors, and related substances). Its possession and use in a sporting context are therefore subject to regulatory prohibition.
