What is IGF-1 LR3?
IGF-1 LR3 (Long R3 Insulin-like Growth Factor 1) is a synthetically engineered analogue of human Insulin-like Growth Factor 1 (IGF-1). It is a modified version of the natural IGF-1 peptide, designed with specific amino acid substitutions that significantly reduce its binding affinity to IGF-binding proteins (IGFBPs). This modification allows the compound to remain active in the bloodstream for a much longer period and to exert stronger and more prolonged biological effects. IGF-1 LR3 is not an approved human or veterinary medicine and is intended solely for research purposes.
Biochemical Properties
IGF-1 LR3 differs from natural IGF-1 in two key ways:
- An arginine substitution at position 3 of the amino acid chain (R3) reduces binding affinity to IGF-binding proteins by up to 1000-fold.
- An additional N-terminal 13-amino acid extension (the Long sequence) substantially increases the half-life of the compound compared to native IGF-1.
The biological half-life of IGF-1 LR3 is approximately 20 to 30 hours, whereas native IGF-1 has a half-life of only a few minutes to hours. These properties make IGF-1 LR3 a valuable tool in cell biology and biomedical basic research.
Mechanism of Action
Like natural IGF-1, IGF-1 LR3 binds to the IGF-1 receptor (IGF-1R), a tyrosine kinase receptor located on the cell surface. Activation of this receptor triggers intracellular signaling cascades, most notably:
- The PI3K/AKT/mTOR pathway, which promotes cell growth, protein synthesis, and cell survival
- The RAS/MAPK pathway, which regulates cell proliferation and differentiation
Due to its reduced binding to IGF-binding proteins, IGF-1 LR3 remains available in its free, biologically active form for longer durations, amplifying its anabolic and anti-apoptotic effects.
Research Applications
In biomedical research, IGF-1 LR3 is used to investigate the role of the IGF-1 signaling pathway in a variety of biological processes. Common research areas include:
- Studying cell growth and proliferation in cell culture models
- Research into muscle cell development (myogenesis)
- Cancer research, particularly the role of IGF-1R in tumor growth and progression

Unauthorized Use and Abuse Potential
Due to its potent anabolic properties, IGF-1 LR3 is misused outside of scientific contexts in competitive and recreational sports to promote muscle growth and accelerate recovery. This use is not medically approved, is not supported by clinical evidence, and carries significant health risks:
- Hypoglycemia (low blood sugar) due to insulin-like activity
- Potential promotion of tumor growth through excessive IGF-1R stimulation
- Acromegaly-like changes, including enlargement of bones and soft tissues
- Disruption of hormonal balance
- Unknown long-term risks, as no clinical studies in humans exist
IGF-1 LR3 is listed on the World Anti-Doping Agency (WADA) Prohibited List and is strictly banned in competitive sports.
Legal Status
IGF-1 LR3 is not approved as a medicinal product in most countries, including Germany and other EU member states. Acquiring, possessing, or using it outside of regulated research settings may have legal consequences. Regulatorily, it is classified as a research chemical and must not be used for therapeutic purposes in humans.
